摘  要:目的研究非毒性分化诱导剂苯了酸钠(phenylbutyrate,NaPB)在体外对人脑胶质细胞瘤BT325细胞的生长、分化的影响。方法应用形态学观察法、MTT分析法、流式细胞仪对在体外经不同浓度的NaPB处理过的BT325细胞进行检测。结果1.电子显微镜观察,在NaPB处理过的BT325细胞中,线粒体、内质网、弹力丝丰富,而在NaPB未处理的细胞中,有大量游离核糖体,上述细胞结构却未能观察到;2.NaPB在2mM时就能明显抑制BT325细胞的生长.并且有时间依赖关系及剂量依赖关系;3.应用流式细胞仪测定发现,经NaPB处理的细胞其S期在11.99%或13.17%.而未经处理的细胞其S期在20.17%。结论NaPB在体外不仅能够抑制人脑胶质细胞瘤的生长,而且能诱导细胞的分化,为NaPB分化治疗胶质瘤提供了实验依据。Objective To study the nontoxic differentiation inducing effect of sodium phenylbutyrate on glioma cells BT325 in vitro. Methods Cell morphology .MTT assay and flow cytometry were used to examine the changes of BT325 cells treated by different concentration of NaPB in vitro.Results 1. Rich mitochondria .rough endoplasmic reticulum and elastic filament were seen in NaPBtreated BT325 cells in electron micrographs. In NaPB-untreated BT325 cells. the above mentioned structures couldn't be observed except numerous freely scattered polyribosomes. 2. BT325 cells were inhibited significantly when the concentration of NaPB reached 2mM. Also NaPB treatment resulted in dose-and time-dependent inhibition of BT325 cells growth. 3. DNA histograms by flow cytometric analysis displayed S phase were 11. 99% or 13. 17% in the NaPB treated BT325 cells and 20. 17 % in the NaPB-untreated BT325 cells. Conclusion NaPB not only inhibits the growth of BT325 cells .but also effectively induces the differentiation of these cells in vitro .providing experimental basis of NaPB differentiation treatment of glioma cells.[
