急性白血病多药耐药基因 MDR1表达及临床意义

摘　　要：①目的　探讨急性白血病（ Ａ Ｌ）多药耐药基因 Ｍ Ｄ Ｒ１ 的表达及临床意义。②方法　采用半定量逆转录聚合酶链反应（ Ｒ Ｔ Ｐ Ｃ Ｒ），对７７ 例 Ａ Ｌ病人及 １０ 例正常人 Ｍ Ｄ Ｒ１ 基因表达进行检测。③结果　７７ 例 Ａ Ｌ病人 Ｍ Ｄ Ｒ１ 阳性率为６４．９％ （５０／７７），其平均表达水平为 ０．２６±０．１９，与对照组比较，差异无显著性（χ２ ＝ ０．０９４，ｔ＝１．５４８， Ｐ＞ ０．０５）；复发组和临床耐药组表达水平明显高于初治组和完全缓解组（ｔ＝ ２．４９，２．４４， Ｐ＜ ０．０５）。④结论 Ａ Ｌ的临床耐药和复发与 Ｍ Ｄ Ｒ１Objective\ To study the expression and clinical significance of multidrug resistance(MDR1) in acute leuke mia. \ Methods\ The expression of MDR1 gene in 77 cases of AL patients and 10 cases of control group was detected with semi quantitative reverse transcription polymerase chain reaction (RT PCR).\ Results\ The positive expression rate (64.9%) and mean expression level (0.26±0.19) of MDR1 in AL patients had no significant difference ( χ 2=0.094, t=1.548,P >0.05,retspectively),compared with those of control group. The expression levels of MDR1 gene in relapse and clinical drug resistant groups were obviously higher than that in early treated and CR groups ( t= 2.49 ,2.26, P <0.05, respectively).\ Conclusion\ The clinical drug resistance and relapse of AL may be related to the overexpression of MDR1 gene.
