摘  要:目的研究培养神经干细胞(NSC)中CNTF,TGF-β1,IGF-1基因表达情况.方法取小鼠胎鼠海马,分离纯化培养NSC,提取总RNA,RT-PCR技术检测CNTF,TGF-β1,IGF mRNA的表达情况.结果培养NSC中各因子mRNA均有表达;CNTF,TGF-β1表达较强,IGF-1表达较弱.结论体外培养的NSC表达CNTF,TGF-β1含量较高,可能是应用其治疗神经疾病的理论基础.Objective To study the expression of neurotrphic factors(NTFs),known as Ciliary Neurotrophic Factor(CNTF),insulin-like growth factor-1(IGF-1),transforming growth factor-beta1(TGF-β1),in the neural stem cells(NSCs)of mice.Methods NSCs were isolated from hippocampus of mice embryo and cultured for 7 days,then were identified by nestin immunohistochemisstry method.Total RNA of NSCs was extracted to determine the expression of mRNA of IGF-1,CNTF,and TGF-β1 by RT-PCR,respectively.Results The mRNA exression for three growth factors were seen in cultured NSC.The expression of CNTF and TGF-β1 was more stronger than IGF-1expression.Conclusions NSCs cultured in vitro can express many neurotrphic factors,the expression levels of CNTF and TGF-β1 are high.This study provides crucial evidence on the application of NSCs in nervous system diseases.
