摘 要:目的探讨不同剂量硫酸镁(MgSO4)预处理及治疗对小鼠全脑缺血再灌注损伤保护作用及可能机制。方法昆明种小鼠,随机分为假手术组、缺血再灌组、MgSO4预处理组和治疗组各分为低、中、高剂量组,预处理组缺血前30 min腹腔注射;治疗组于再灌后即刻腹腔注射。观察病理形态学变化,检测脑组织一氧化氮(NO)含量和总一氧化氮合酶(T-NOS)、诱导型NOS(iNOS)活性,ATP酶的活性。结果 MgSO4预处理和治疗各组脑组织缺血再灌注损伤病理学改变明显轻于再灌组。假手术组及MgSO4预处理和治疗各组脑组织NO含量和NOS、iNOS活性均低于再灌组(P<0.05,P<0.01)。脑组织钠钾ATP酶及钙镁ATP酶的活性假手术组及MgSO4预处理和治疗各组均高于缺血再灌组(P<0.05)。结论 MgSO4预处理和治疗均可能通过降低NOS及iNOS活性及NO含量,增加ATP酶的活性,减轻神经元损伤程度,对缺血再灌注脑损伤具有保护作用。Objective To explore the protective effect and possible mechanism of different doses of magnesium sulfate(MgSO4) pretreatment and treatment for global cerebral ischemia reperfusion(I/R) injury in mice.Methods The mice were randomly divided into sham,I/R and pretreatment,low,medium and high doses of MgSO4 treatment groups.Mice in pretreatment group were injected intraperitoneally with MgSO4 before 30 min ischemia.Mice in treatment group were injected intraperitoneally immediately after reperfusion.The pathologic change was observed.The content of nitric oxide(NO),the activity of total nitric oxide synthase(T-NOS) and inducible nitric oxide synthase(iNOS) and ATPase of cerebral tissue were also detected.Results The pathologic change of ischemic impairment in MgSO4 pretreatment and treatment groups were significantly lighter than that of I/R group.The content of NO and the activities of T-NOS and iNOS of cerebral tissue in the sham,MgSO4 pretreatment and treatment groups were all significantly lower than those of I/R group(P0.05,P0.01).The activity of Na+-K+-ATPase and Ca2+-Mg2+-ATPase of cerebral tissue were all higher than those of I/R group(P0.05).Conclusions MgSO4 pretreatment and treatment may reduce the degree of neuron injury and have protective effect on I/R cerebral injury,which may be associated with the decrease of the content of NO and the activities of NOS,iNOS and the increase of the activity of ATPase.
