β-catenin和MMP-7在异常腺窝病灶中的表达差异的初步探讨

摘　　要：目的检测β-catenin及MMP-7在Wistar鼠大肠癌模型异常腺窝病灶中的表达差异,探讨其与大肠肿瘤的相关性及其发生途径。方法 20只Wistar鼠给予二甲肼经典造模处理,于24周处死,取其大肠组织进行病理和免疫组化检查。结果发现两种不同病理类型的ACF:hACF和dACF,两者β-catenin的表达异常率分别为4.84%和100%,在MMP-7的阳性表达率分别为7.87%和81.82%,而肿瘤的β-catenin的表达异常率和MMP-7的阳性表达率均为100%。dACF与肿瘤在两个癌基因表达率的差异均没有统计学意义。结论单纯增生型ACF与肿瘤发生无明显相关,非典型性增生型ACF与肿瘤发生关系密切,并且其发生可能遵循Wnt途径。【Objective】 To evaluate the relationship between Aberrant Crypt Foci（ACF） and colorectal tumor and the ACF＇s growth signal way by examining the different expressions of β-catenin and MMP-7 in ACF.【Methods】 20 Wistar rats were injected in DMH in a dose of 15mg per kg,once a week,continuing 18 weeks,then killed in week 24,the colons were removed to pathology and the immunohistochemical inspection.【Result】 There were two types of ACF,hyperplasia ACF and dysplasia ACF.The difference abnormal expressed rate of β-catenin in the two types of ACF respectively was 4.84% and 100%（P =0.000）,the MMP-7 expression rate was 100%.【Conclusion】 Dysplasia ACF related to colorectal tumor,in contrast,hyperplasia ACF is not.Dysplasia ACF occurs and possibly follows the Wnt signal way.
