摘  要:目的研究氧化槐定碱(oxysophoridine,OSR)对福尔马林致痛大鼠脊髓背角氨基酸含量的影响。方法实验采用Wist-ar大鼠,OSR(1 000,500,250 mg.kg-1,ip)1 h后右后足跖掌部皮下注射福尔马林(5%,100μL)致痛,30 min后取脊髓L4-6节段背角组织,采用氨基酸自动分析仪检测脊髓组织氨基酸含量。结果 OSR(1 000,500 mg.kg-1)可使福尔马林致痛大鼠脊髓L4-6节段背角组织中谷氨酸含量明显下降(P<0.05),OSR(1 000 mg.kg-1)还能明显增加γ-氨基丁酸和甘氨酸含量(P<0.05,P<0.01)。结论 OSR能降低福尔马林致痛大鼠脊髓L4-6节段谷氨酸含量并升高γ-氨基丁酸和甘氨酸含量,此作用与OSR的镇痛作用有关。OBJECTIVE To study the effect of oxysophoridine (OSB) on the amino acid content in the tissue of L4-6 section in the spinal dorsal horn of rats with formalin induced-pain. METHODS Forty Wistar rats (265 ± 15 ) g were randomly divided into 5 groups with 8 in each group i. e. control group, model group and OSR ( 1 000, 500,250 mg . kg-1 respectively) groups. Pain stimulation was produced by subcutaneous injection of 5% formalin solution 100 μL into the plantar region of right hind paw in model group and OSR groups, while the control group was injected 100 μL normal saline (NS). 1 h before pain stimulation, NS (5 mL . kg-1) ,control group and model group were administered through intraperitoneal injection (ip) ; OSR groups (1 000, 500, 250 mg . kg-1) was administered OSR solution (5 mL . kg-1 ) through ip. All rats were killed at 30 min after injecting 5% formalin solution into planta, and the tissue of IA-6 section in the spinal dorsal horn was taken. The content of amino acid in the tissue of LA-6 section in the spinal dorsal horn was assayed by amino acid autoanalyzer. RESULTS The amino acid contents (glutamate, Glu) in the tissue of IA-6 section of the spinal dorsal horn in rats with formalin induced-pain decreased significantly in OSR 1 000, 500 mg . kg - 1 groups ( P 〈 0. 05 ), the contents of γ-aminobutyric acid (GABA) and glycine (Gly) increased significantly in OSR 1 000 mg . kg-1 group (P 〈 0. 05, P 〈0. 01 ). CONCLUSION OSR can decrease glutamate content and increase γ-aminobutyric acid content in the tissue of IA-6 section of the spinal dorsal horn in rats formalin induced-pain with, which may be related to the analgesic effect of OSR.
