摘 要:[目的]明确高分子亚基Bx7OE在6个品种中的分布,研究其对小麦品质影响,帮助品质性状改良。[方法]利用已有Bx7OE特异性标记,对津强1号、辽春10号及其杂交育成的5个品种进行检测,采用SDS-PAGE方法,获得7个品种高分子量亚基分布数据,并进行品质分析。[结果]7个供试品种中,有4个品种扩增出447 bp的片段,分别是津强1号、津强2号、津强5号和津强6号,说明里面含有7OE亚基。SDS-PAGE数据表明,这7个品种亚基组成为在Glu-A1位点除津强1号为2*亚基外,其余均为1亚基,在Glu-B1和Glu-D1位点均为7+8亚基和5+10亚基。通过品质数据分析,Bx7OE亚基对小麦粉质指标有明显的正面影响。[结论]STS标记特异性较好,Bx7OE亚基对改良小麦品质作用较大,可用于中国小麦品质改良。[Objective] This study was to characterize Bx7OE gene in 7 wheat cultivars and its impacts on wheat quality improvement.[Method] Employing Bx7OE-specific marker,distribution of Bx7OE gene in seven wheat varieties of Jinqiang No.1,Liaochun No.10 and their hybrids was detected to get the high molecular weight glutenin subunit(HMW-GS) compositions using sodium-dodecyl-sulfate polyacrylamide gel electrophoresis(SDS-PAGE),and the data obtained were further used for quality analysis.[Result] Of the seven varieties tested,four of Jinqiang No.1,Jinqiang No.2,Jinqiang No.5 and Jinqiang No.6 produced a 447-bp PCR fragment from PCR amplification with TaBAC1215C06-F517/R964 as primers,suggesting that these varieties harbor 7OE subunit.From the data of SDS-PAGE,at Glu-A1 locus,except Jinqiang No.1 with subunit 2*,all the others harbor subunit 1;at Glu-B1 locus and Glu-D1 locus,all harbor subunits 7+8 and 5+10,respectively.At the Glu-B1 locus,subunits Bx(7OE+8) showed significantly better effects on quality parameters than subunit Bx(7+8).[Conclusion] Thanks to the good specificity of STS markers,Bx7OE subunit,which has better impacts on the improvement of wheat quality,could be used for the genetic improvement of Chinese wheat quality.
