摘 要:目的:探讨层粘素 P11 活性片段对胆管癌细胞浸润过程的影响及可行的抗浸润治疗途径。方法:以层粘素 P11 片段处理 Q B C939 胆管癌细胞,观察胆管癌细胞分泌Ⅳ型胶原酶、尿激酶型纤维蛋白溶酶原激活物的变化,并利用人羊膜浸润培养系统观察胆管癌细胞浸润能力的改变。结果:与层粘素整体分子相反,层粘素 P11 片段能够显著抑制胆管癌细胞分泌Ⅳ型胶原酶和体外浸润能力,但对尿激酶型纤维蛋白溶酶原激活物的分泌无明显影响。结论:层粘素可能通过胆管癌细胞膜细胞表面 L N 受体刺激Ⅳ型胶原酶分泌,介导胆管癌细胞浸润和转移过程。而浸润和转移过程中另一重要蛋白酶尿激酶型纤维蛋白溶酶原激活物的分泌调节,可能通过其它途径实现。研究阻断层粘素与层粘素受体结合的治疗方法,可能在未来临床防治肿瘤浸润和转移治疗中具有光明前景。Objective: To study the effects of laminin segment, P11 amide, on the invasive potential of malignant biliary cells and look for a possible anti invasive agent. Methods: After QBC 939 cells of biliary malignancy were cultured with P11 amide, the changes of the secretion of type Ⅳ collagenase and urokinase type plasminogen activator were determined. In a human amnion infiltration culture system, the invasive potential of QBC 939 cells before and after the treatment of P11 amide were observed. Results: Laminin segment or P11 amide, contrary to laminin, inhibited significantly the secretion of type Ⅳ collagenase, showed no effect on the secretion of urokinase type plasminogen activator and suppressed the invasive potential of the treated QBC 939 cells. Conclusion: Laminin induces the secretion of type Ⅳ collagenase in malignant biliary cells and mediates their malignant invasion through its combination with its receptors on the surface of malignant biliary cells. The employment of P11 amide blocks the combination between laminin and its receptors which might serve as an anti invasive agent for biliary cancer.
