摘 要:目的 研究蛋白激酶C(protein kinase C,PKC)信息通道在阻塞性黄疸机体发病机理中的作用。 方法 从51 例阻塞性黄疸患者和16 例正常对照者的外周静脉血中分离和纯化淋巴细胞胞浆及胞膜部分蛋白激酶C,然后采用同位素γ32PATP催化活性测定法检测它们的活性。 结果 (1)阻塞性黄疸患者外周血淋巴细胞蛋白激酶C的总活性较正常健康人明显增强( P< 0-01) ,且胞膜部分蛋白激酶C活性占总活性的百分值较正常组也明显升高( P< 0-05)。(2)阻塞性黄疸患者淋巴细胞蛋白激酶C的活性与血清黄疸的程度(TBIL) 显著正相关(r= 0-67,P< 0-01),与血清可溶性白细胞介素2 受体(soluteinterleukin2,sIL2R) 浓度显著正相关(r= 0-58 ,P< 0-01)。 结论 蛋白激酶C信息通道的激活与黄疸程度有关,且可能参与了阻塞性黄疸时T淋巴细胞的活化。Objective To study the roles of protein kinase C (PKC) signal pathway in the pathogenesis of patients with obstructive jaundice. Methods PKC from cytosolic and membrane fractions of peripheral blood lymphocytes (PBL) was isolated and purified, its activities were determined by radioactive isotope γ 32 P ATP catalyzing assay. Results Fifty one patients with abstructive jaundice and sixteen normal controls were enrolled in this study. (1) The total PKC activities in PBL in patients with obstructive jaundice siginificantly increased as compared to those in normal subjects ( P <0 01). Moreover, the membrane PKC activities and their percentages of the total PKC activities were higher in patients with obstructive jaundice than in normal subjects ( P <0 05).(2) The total PKC activities in PBL was in positive correlation with the levels of solute IL 2 receptor (sIL 2R) ( r =0 67, P <0 01) and the degree of jaundice (T BIL) ( r =0 58, P <0 01) in serum. Conclusions The activities of PKC signal pathway related with the degree of jaurdice,it may be implicated in the activation of T lymphocytes. PKC signal pathway may be important in the immune regulation and the assessment of the condition in patients of obstructive jaundice.
