摘 要:目的:确定新城鸡瘟病毒Losota系(NDVL)能否诱导小鼠腹腔巨噬细胞(PEMφ)产生一氧化氮(NO)以及该NO与小鼠PEMφ介导的细胞毒作用是否有依赖关系。方法:采用Gries、FACS分析和噻唑蓝(MTT)方法。结果:NDVL与PEMφ作用2h后就可以稳定地吸附在PEMφ表面,吸附率达80%左右;收集NDVL与PEMφ作用24h上清,发现有NO的释放,释放量与阳性对照组(BCGLPSPEMφ上清)的释放量接近;采用MTT法测定NDVLPEMφ对H22肝癌细胞、P815肥大细胞瘤、Raji细胞和K562白血病细胞的细胞毒活性(效/靶比为20∶1),发现NDVLPEMφ对这些靶细胞具有明显的杀伤活性,且该活性与NDVLPEMφ诱导NO之间有一定的依赖性。结论:新城鸡瘟病毒Losota(NDVL)能诱导小鼠巨噬细胞产生和释放一氧化氮。Objective:Determine if NDV L can induce mouse PEMφ releasing Nitric Oxide (NO) and the relationship between the NO and the cytotoxic activity of PEMφ. Methods:Used Griess FACS and MTT analysis. Results:NDV L could absorb steadily on the surface of PEMφ after infecting PEMφ for 2 h and absorption ratio was about 80 percent. When detecting the supernatant liquid which NDV L infected PEMφ for 24 h found that the quantities of NO was almost equal to the positive control group (BCG LPS PEMφ); While using MTT assay also found that the cytotoxic activities of NDV L PEMφ against H 22 , P 815 , K 562 and Raji targets was much higher than the ones of control group and there were some dependent relationship between NO and the cytotoxic activities of PEMφ. Conclusion:Newcastle Disease Virus can induce macrophages producing Nitric Oxide (NO) and there was relationship between the NO and the cytotoxic activities of PEMφ.
