均匀设计法优化消癥方处方配比的研究

摘　　要：目的应用均匀设计方法实现消癥方的配比优化。方法将小鼠荷瘤后随机分为12组,其中7组按照均匀设计要求给予不同配比的重楼总皂苷、吴茱萸总碱及钩藤总碱,另外5组分别为中药单药组、环磷酰胺组和空白对照组各1组,连续灌胃给药14天后,次日处死。应用均匀设计方法,以抑瘤率为疗效指标,对数据进行处理。结果由均匀设计方法得到抑瘤率方程(Y=0.851 78-0.014 92×X2-0.000 390 07×X1X2-0.000 878 39×X1X3),其中当组方中重楼总皂苷为37.5mg、钩藤总碱为12.5mg时抑瘤效果最好,可达44%。单药钩藤总碱组小鼠体重增加最明显,由给药前的(25.12±2.36)g增至(33.06±2.47)g,与空白对照组小鼠体重相比差异有统计学意义(P<0.05)。结论均匀设计法对探索中药组方配伍规律是一个大胆的探索,经过现代数理统计法确定的处方用量的优化配比,可以为中药方剂的现代化研究提供依据。Objective To optimize the proportion of the prescription of Xiaozheng by uniform design method.Methods The mice were divided into twelve groups,including seven groups treated with the prescriptions which contained rhizoma paridis total saponins,evodiamine and rhymchophylla total alkaloids with different proportion,three groups treated with single traditional Chinese medicine mentioned before,cyclophosphamide group and distilled water group.On the second day,each group was treated with relevant medicinal for fourteen days.The proportion was fixed using uniform design method.The inhibition rates of different formulations of prescription of Xiaozheng on murine xenograft was compared.Results The regression equation was Y=0.851 78-0.014 92×X2-0.000 390 07×X1X2-0.000 878 39×X1X3.The best proportion of the prescription for rhizoma paridis total saponins was 37.5mg and rhymchophylla total alkaloids was 12.5mg,and tumor inhibitory ration was 44%.The body weight of murine were increased significantly in the rhymchophylla total alkaloids group,from（25.12±2.36）g to（33.06±2.47）g,which was higher than that of distilled water group（P0.05）.Conclusion Compatibility of medicines in prescription is the soul of traditional Chinese medicine theory.It＇s a courageous method to explore the law of prescription by the uniform design method.The optimized prescription could achieve the best effect.The optimized proportion of prescription using modern mathematical statistics method can promote the modernization of traditional Chinese medical research.
