摘  要:目的:探讨血管生成抑制剂Suramin对实验性大鼠乳腺癌的化学预防作用。方法:40只SD雌性大鼠,随机分为4组:A组12只,腹腔注射甲基亚硝脲(MNU);B组8只,腹腔注射MNU,在导管内癌(DCIS)生成高峰前皮下注射Suramin;C组8只,腹腔注射MNU,在DCIS生成高峰后皮下注射Suramin;D组12只,腹腔注射MNU,在DCIS生成高峰前皮下注射他莫西芬(TAM)。观察各组大鼠肿瘤生长情况,记录肿瘤大小和数量;在鼠龄98 d时,取各组大鼠乳腺组织行常规HE染色病理学检查,应用SP免疫组化法检测构建大鼠乳腺IDP、DCIS和浸润癌(IDC)中血管内皮生长因子(VEGF)表达及MVD。结果:B组和C组的IDC发生率均较A组明显下降,差异有统计学意义(P<0.05),而两组的IDC发生率与D组比较差异无统计学意义,P>0.05。B组和C组平均肿瘤体积较A组和D组均有明显下降,差异有统计学意义,P=0.01。A组、B组、和C组的IDC中,MVD计数依次为79.9±8.6、44.4±6.2和59.1±7.1,各组所有病变的平均MVD计数依次为61.3±2.4、29.6±1.2和26.1±3.3,B组和C组所有病变的平均MVD均较A组明显下降,差异有统计学意义,P=0.005和P=0.01。IDP、DCIS和IDC中的VEGF表达率分别为30.4%、42.9%和83.1%,全部IDP、DCIS和IDC中的MVD计数分别为17.3±3.4、30.4±3.6和68.2±8.6,VEGF与MVD呈正相关。结论:Suramin在乳腺癌的发生发展过程中有明显的抑制作用,对乳腺癌的发生发展有一定的化学预防作用。OBJECTIVE: To investigate the chemopreventive effect of angiogenrsis inhibitor Suramin on induced breast cancer in rats. METHODS=A total of 40 SD rats were randomly divided into four groups= 12 rats in group A(negative control group), injected with MNU i. p. only; 8 rats in group B(test group 1), injected with MNU i. p and injected with Suramin i.h. before development of DCIS; 8 rats in Group C(test group 2), injected with MNU i. p and injected with Suramin i.h. after development of DCIS; Group D(positive control group), injected with MNU i. p and injected with TAM i. h. before development of DCIS. The diameter and number of the tumors were observed, all rats were killed at 98 days of age, then evaluated the breast lesions by microscopy and detected the expression of VEGF and MVD in IDP, DCIS and IDC by SP. RESULTS: The rates of IDC in group B and C were lower than those in group A, and there was a statistical significance between each test group and negative control group (P〈0. 05). But between each test group and positive control group were not significant difference (P〉0. 05). The average volume of tumors in group B and C were less than group A and D, and there was a statistical significance between each test group and each control group (P= 0.01). The MVD of case with IDC in groupA, B and C were 79. 9±8. 6, 44. 4±6. 2, and 59.1±7.1 respectively, and MVD of case with all pathological changes in the three groups were 61.3 ± 2. 4, 29.6±1.2 and 26.1±3.3. There was a statistical significance between each test group and negative control group (P=0. 005 and P=0.01). The positive rate of VEGF in all IDP, DCIS and IDC was 30.4%, 42.9% and 83.1% respectively, and MVD was 17.3±3. 4, 30. 4±3. 6 and 68. 2±8. 6 respectively. There was a significant correlation between MVD and expression of VEGF. CONCLUSIONS: Suramin can decrease the neoplasitc lesions rate and volume of tumors obviously in MNU-indueed rats.
