摘 要:目的探讨肥胖大鼠心肌细胞内钙调节机制。方法 60只健康雄性SD大鼠高脂饲料喂饲10周后,筛选出肥胖易感(OP)大鼠,基础组大鼠和肥胖抵抗(OR)大鼠,每组15只。测量各组大鼠体脂肪含量,检测血脂水平。采用酶法分离心肌细胞,Fluo-3/AM负载后采用激光共聚焦扫描显微镜观察各组大鼠心肌[Ca2+]i对KCl除极及咖啡因诱导的反性。结果与基础组大鼠和OR大鼠比较,OP大鼠睾周脂肪、肾周脂肪、网膜脂肪、体脂比升高(P<0.05),血清总胆固醇、三酰甘油和低密度脂蛋白水平升高(P<0.05),心室肌[Ca2+]i的升高幅度降低(P<0.05)。结论肥胖心肌细胞[Ca2+]i在KCl除极后及咖啡因诱导后升高幅度发生改变,可能是肥胖者心律失常发生机制之一。Objective To study the intracellular calcium([Ca2+]i) in myocardial cell of obesity rats induced by high-fat diet.Methods Male Sprague-Dawley(SD) rats were divided into obesity resistant(OR,n=15),normal(Nor,n=15) and obesity prone(OP,n=15) group after fed with high-fat diet for 10 weeks.Their body fat and serum lipids were measured.Myocardial cells were isolated with Langendorff perfusion and [Ca2+]i was measured with calcium indicator Fluo-3/AM and laser scanning confocal microscope after KCl depolarization and caffeine-induced.Results Compared with those in Nor and OR rats,the epididymal fat,perirenal fat,omental fat and body fat increased in OP rats(P0.05),as well as the the level of total cholesterol,triglyceride and low density lipoprotein(P0.05);the vary of [Ca2+]i elevation and restoration were lower(P0.05).Conclusion The vary of [Ca2+]i elevation decreases in OP rats after KCl depolarization and caffeine-induced,that may associated with arrhythmia in obesity rats.
