肝癌中肝细胞生长因子的表达及意义

摘　　要：目的:研究肝细胞生长因子(HGF)在肝癌患者组织及血浆中的表达,探讨HGF在肝癌发生、发展中所起的作用。方法:对肝癌及慢性乙型肝炎患者进行调查,并且设立正常对照组,应用酶联免疫吸附法检测各组血浆HGF水平;应用半定量RT-PCR法和免疫组织化学法分别检测各组肝组织中的HGF mRNA及HGF蛋白的表达。结果:(1)PCR产物行半定量分析,各组数据两两比较差异均有统计学意义(P<0.01),肝炎组和肝癌组中HGFmRNA的表达均显著高于正常肝组织,且在肝癌组中的表达显著高于肝炎组。(2)免疫组化法检测肝癌组织中HGF阳性染色位于细胞胞浆,HGF的表达阳性率97.1%。肝炎组织中HGF的表达阳性率51.4%,正常肝组织阳性率0%。且肝癌组织中HGF的表达高于肝炎组织,差异有统计学意义(P<0.01)。(3)ELISA检测肝癌HGF与其它两组间比较差异有统计学意义(P<0.01),其中肝癌组血浆中HGF高于肝炎组,而对照组最低。结论:HGF在肝癌组织及血浆中呈高表达,可能与肝癌的发生、发展有关。Objective：To study the expression of hepatocyte growth factor（HGF） in the serum and tissues in patients with hepatic carcinoma,and to explore the role of HGF in the tumorigenesis and progression of human hepatic carcinoma.Method： Study the patients with hepatic carcinoma and hepatitis were studied,and the normal control group was established,their plasma HGF concentrations were measured by enzyme-linked immunosorbent assay（ELISA）;the expression of HGF mRNA and protein were detected in their liver tissue of them by using semi-quantitative reverse transcription-polymerase chain reaction（RT-PCR） and immunohistochemistry respectively.Results：The plasma HGF concentrations in patients with hepatic carcinoma was higher than those in the hepatitis and normal control group（P0.01）.The expression level of HGF in hepatic carcinoma was higher than those in the hepatitis and normal hepatic tissue（P0.01）.Conclusion：The expression of HGF in the serum and tissues in patients with hepatic carcinoma was higher.HGF may be involved in the tumorlgenesis and progression of hepatic carcinoma.
