摘 要:本文研究使用高效萃取体系,建立反相高效液相色谱(RP-HPLC)同时测定食品中对位红、苏丹红Ⅰ~Ⅳ号及苏丹红7B。样品用甲醇-乙腈-丙酮萃取液提取,采用聚硅烷C18色谱柱(Shiseido C18 150mm×4.6mmi.d.,5μm)分离以水-乙腈为流动相,流速1.0mL/min,等度洗脱;用紫外一可见光二极管阵列检测器测定,检测波长为500nm。研究结果表明该方法对苏丹红、对位红的最低检测限为0.1mg/kg,标准曲线的线性相关系数线(r=0.9998),在0.5-5.0μg/mL添加水平,加标回收率为85%-~106%,RSD为2.14%0.82%。该方法涉及的化学试剂少,设备少,操作简便,结果准确,是食品中对位红、苏丹红类染料的快速有效检测方法。A method was developed for the determination of Para Red and Sudan dyes in food by reversed phase-high performance liquid chromatography (RP-HPLC) with a new simple extract system. Sample was extracted from food with methanol-acetonitrile-acetone. The analysis was performed on a Shiseido C18 column(150 mm×4.6mm i.d., 5μm), with water-acetonitrile as the mobile phase at a flow rate of 1.0mL/min. Detection was carded out at the wavelength of 500nm and confirmed by spectra. All of the six compounds demonstrated good linear relationship (r=0.9998). The limit of detection (LOD) (spiked at the levels of 0.5-5.0 μg/mL) was 0.1 mg/kg. The average recoveries for all six dyes were ranged from 85%-106%, and the relative standard deviations were form 2.14%.-4.82%. The method is sensitive, reliable and can be applied for the analysis of Para Red and Sudan dyes in food.
