摘 要:目的:观察氧化应激对心肌成纤维细胞(CFb)衰老的影响以及3'-大豆苷元磺酸钠(DSS)的干预作用,初步探讨3'-大豆苷元磺酸钠抗心脏老化过程中心肌纤维化的机制。方法:差速贴壁法培养SD乳鼠心肌成纤维细胞,MTT法检测细胞增殖能力,硝酸还原酶法检测细胞上清液NO含量,采用免疫细胞化学染色检测心肌成纤维细胞的-平滑肌肌动蛋白(-SMA)表达,用吖啶橙荧光染色检测细胞的衰老。结果:在H2O2刺激下,心肌成纤维细胞增殖率降低,-SMA免疫化学染色出现明显强阳性,与正常组比较有显著性差异(P<0.01),NO含量与正常组比较明显降低(P<0.01),3'-大豆苷元磺酸钠3种浓度均具有延缓H2O2诱导的以上效应,与H2O2刺激组比较有显著性差异(P<0.05;P<0.01)。结论:3'-大豆苷元磺酸钠通过抑制氧化应激加速心肌成纤维细胞的衰老,恢复心肌成纤维细胞正常增殖能力,其作用机制可能是通过影响NO系统以及受体机制来抑制心肌成纤维细胞表型转化,这也可能是3'-大豆苷元磺酸钠抗心肌肥厚的机制之一。Objective:Observation of oxidative stress on cardiac fibroblasts(CFb) senescence and the intervention of 3′-daidzein sulfonate sodium(DSS).To explore the mechanism of 3′-daidzein sulfonate sodium on Anti-aging process of myocardial fibrosis in the heart.Methods:Cultured cardiac fibroblasts of SD neonatal rat by differential attachment method,detected cell proliferation by MTT method and NO content of cell supernatant by nitrate reductase method.Detected the expression of-smooth muscle actin(α-SMA) of cardiac fibroblasts by immunocytochemical staining method and cell senescence using acridine orange staining.Results:In H2O2 stimulated the proliferation rate of cardiac fibroblast decreased,α-SMA immunohistochemical staining showed strong positive.Compared with the normal group there was significant difference(P0.01).NO content was significantly lower than the normal group(P0.01).3 concentrations of 3′-daidzein sulfonate sodium were more than delaying effects which were induced by H2O2.There were significant difference compared with H2O2 stimulated groups(P0.05;P0.01).Conclusion:3′-daidzein sulfonate sodium accelerated aging cardiac fibroblasts by inhibiting the oxidative stress and restore the normal proliferation of cardiac fibroblasts.Its mechanism may inhibit the cardiac fibroblasts phenotype through influencing the NO system and the receptor mechanism.It may also be one of the mechanisms of 3′-daidzein sulfonate sodium against cardiac hypertrophy.
