摘  要:目的:体外观察谷氨酰胺(GLN)联合5-氟尿嘧啶(5-Fu)对人胃癌细胞株MGC-803的抑制作用及对细胞周期的影响,初步探讨GLN联合5-Fu用于胃癌治疗的有效、合理方案。方法:采用四甲基偶氮唑盐(MTT)法检测药物对细胞的抑制作用,应用中效原理判定两药合用的效果。流式细胞仪检测细胞周期分布。结果:GLN和5-Fu的相互作用关系与药物剂量有关:中小剂量时呈拮抗作用,较大剂量时呈协同作用。5-Fu对细胞周期影响较小,仅表现为轻度S期(24~48h)及G0/G1期(72h)阻滞作用。联合应用GLN后,仍对细胞周期影响较小。5-Fu作用时间、药物浓度、是否联合GLN,均对细胞增殖指数(PI)影响较小。结论:GLN并不能通过影响细胞PI而影响5-Fu抗肿瘤作用。对于胃癌化疗的患者,联合应用较大剂量的5-Fu和GLN,可能是增强疗效的合理用药方案。Objective:To evaluate the growth inhibiting effects and changes in cell cycle distribution caused by combining glutamine(GLN)with 5-fluorouracil(5-Fu) on human gastric carcinoma cell line MGC-803 in vitro and to explore the effective and reasonable scheme for gstric cancer treatment.Methods:The effects of GLN and 5-Fu were measured by MTT assay,and the interaction between the two agents was evaluated by the median-effect principle.Flow cytometry was used for cell cycle analysis.Results:The interaction of these two agents was related to drug concentrations:high-dose combination results in synergistic cytotoxic effects and low-dose combination results in antagonistic cytotoxic effects.5-Fu could hardly influence cell distribution,only showing mild S-phase arrest(24-48h) and mild G0/G1 phase arrest(72h).Moreover the effection on cell distribution was still feeble by combining GLN with 5-Fu.Regardless of durg duration,durg concengtion,and whether combining with GLN,5-Fu could hardly change the cell proliferation index.Conclusion:GLN could not influence the antitumor effection of 5-Fu by means of increasing proliferation index.The combination of high-dose 5-Fu and GLN may achieve better efficacy for gastric cancer treatment.
