摘  要:目的探讨细胞外基质磷酸化糖蛋白(MEPE)对人牙髓细胞(HDPCs)黏附、伸展、增殖的影响。方法常规培养获得人牙髓细胞,各实验组中MEPEs的浓度分别为0.01,0.1,1,10,100μg/L以不加MEPE作为空白对照组。细胞计数法测定细胞的黏附能力;通过计数高倍视野中伸展的细胞数,计算骨髓基质细胞在不同时间的伸展率;MTT法检测各组细胞的增殖活性。结果体外培养的人牙髓细胞生长良好;各实验组间及与对照组比较,100μg/L组对细胞的黏附性的影响有差异;各组细胞的伸展率无差异;MEPE对人牙髓细胞有较明显的促增殖作用。结论 MEPE对体外培养的人牙髓细胞的伸展率无影响,但可明显促进其黏附性和增殖。Objective To evaluate the effects of Matrix extracellular phosphoglycoprotein(MEPE) on the attachment,spreading and proliferation of human dental pulp cells(HDPCs).Methods HDPCs were conventional cultured.MEPEs was added into medium in several concentrations(0.01,0.1,1,10,100μg/L) as experimental groups.HDPCs were cultured without MEPE as control group.Attachment ability of MEPEs was detected by counting cell number.Cell spreading rates were performed at various culture times by analysis of micrographs taken at predetermined sites of each wells.Cell proliferation rates were assessed by MTT assay.Results It was shown that MEPEs were cultured successfully in vitro.There was no significant change between the control group and experimental groups in cell spreading rate.However,the cell attachment was significantly in 100μg/L group.The cell proliferation of MEPEs was significantly stimulated by MEPEs in a dose-and time-dependent manner.Conclusion MEPEs could promote the attachment and proliferating ability of human MEPEs,but have no effects on its spreading.
