摘 要:目的:探讨标记抗人肝癌单克隆抗体超顺磁性氧化铁粒子(McAb-SPIO)对荷瘤裸鼠模型的诊断作用。材料和方法:用过碘酸盐法将Hab-18 抗人肝癌单抗标记到SPIO粒子;建立裸鼠载瘤模型(n=12 只)。结果:实验组(n= 8)从尾静脉注入0.3 m g Fe/20g 的McAb-SPIO,给药后24 小时成像观察到裸鼠瘤结均有选择性信号下降,对照组(n= 4)在注射LgG-SPIO后未见明显信号改变。结论:McAb-SPIO磁共振免疫成像对肝癌的早期定性诊断有较高价值。Purpose:To explore The efficacy of MR antibody imaging with superparamagnetic iron oxide particles (SPIO) coated by Monoclonal Antibodies (McAb) in the diagnosis of nude mice carrying liver tumors. Materials and Methods: McAb against human hepatocellular carcinoma coating SPIO was achieved with periodate method. 12 nude mice with the tumor were established by subcutaneously injection of QGY 7723 hepatocellular carcinoma cell line. The tumor grew up about 0.5 1.0cm in diameter was harvested for further MR study.Results: After i.v. injection of 0.3 mg/20 g body weight of McAb SPIO, heterogeneous specific signal reduction was noticed in experimental group (n=8), while no obvious signal intensity changes in the control group (n=4) were founded after injection of nonspecific lgG SPIO . Conclusion: McAb SPIO enhanced MR imaging is a promising way for early diagnosis of tiny hepaticellular carcinoma nodules.
