摘 要:目的分析高同质性宫颈鳞癌细胞与正常宫颈细胞蛋白质的表达差异,以期获取宫颈鳞癌的分子标志物。方法利用激光捕获显微切割(LCM)技术分离获取宫颈鳞癌和正常宫颈组织细胞,双向凝胶电泳和质谱法检测两者表达的差异蛋白。结果双向凝胶电泳得到20个差异蛋白点,质谱鉴定显示,其中ETS1、HSP60、Bcl-2、C-myc、TIF1-β等5种蛋白在宫颈鳞癌细胞中表达明显增高(P均<0.01)。结论应用LCM技术可以成功获取同质宫颈癌细胞和正常细胞;宫颈鳞癌和正常宫颈细胞的2-DE蛋白质图谱具有明显的差异表达,该表达差异蛋白可望作为宫颈鳞癌的分子标志物。Objective To analyse the differences of protein components in high homogeneity cervical squamous cell carcinoma and normal cervical,to obtain molecular markers which could identify cervical squamous cell carcinoma.Methods Homogeneous cells of cervical cancer and normal cervical tissue were collected by LCM,and subsequently were analyzed with 2D-DIGE.The interest protein spots were picked up and digested by trypsin,and then identified with matrix-assisted laser desorption and ionization time-of-flight mass spectrometry(MALDI-TOF-MS).Results Significant expression changes were seen with 20 protein spots resulting in identification of 5 unique proteins,including ETS1,HSP60,Bcl-2,C-myc and TIF1-β that uniquely distinguish normal cervical tissue.Conclusions Homogeneous cells from cervical cancer and normal cervical tissue have been successfully collected with LCM.Significant difference of 2D-DIGE mapping between cervical cancer and normal cervical tissue was observed.Novel markers described here may have potential applications for increasing the predictive value of current screening methods and therapeutic interventions in cervical cancer.
