膀胱移行细胞癌组织中has-miR-200b、E-cadherin的表达变化及意义

摘　　要：目的检测膀胱移行细胞癌(BTCC)组织中hsa-miR-200b基因、E-cadherin蛋白的表达变化,并探讨其意义。方法采用免疫组化法检测58例BTCC患者肿瘤组织中的E-cadherin蛋白,用实时定量PCR法检测其中的hsa-miR-200b基因。结果 BTCC组织中E-cadherin表达缺失率为25.86%(15/58),hsa-miR-200b表达量为36.27±34.2;BTCC组织中E-cadherin蛋白、hsa-miR-200b基因表达均与BTCC病理分级有关(P均<0.05)。E-cadherin正常表达组的hsa-miR-200b基因表达量为2.63±0.39,与E-cadherin表达缺失组的0.73±0.39相比明显增高(P<0.001)。结论 BTCC组织中hsa-miR-200b、E-cadherin蛋白的表达上调,且二者的表达有相关性。二者可能在在BTCC发生发展过程中起重要作用。Objective To study the expression of E-cadherin protein and hsa-miR-200b gene in bladder transitional cell carcinoma（BTCC） tissue.Methods E-cadherin of 58 cases of nasopharyngeal carcinoma was detected by immunohistochemisty and hsa-miR-200b gene was by RT-PCR.Results Loss of expression of E-cadherin was 25.86%（15/58）.The relative expression value of hsa-miR-200b was 36.27±34.23.Both E-cadherin and hsa-miR-200b were associated with staging of BTCC.Compare with the loss of expression group,the normal E-cadherin expression group has higher expression of hsa-miR-200b（P0.001）,the relative expression value was 2.63±0.39 and 0.73±0.39 respectively.Conclusions The expression of hsa-miR-200b gene is associated with E-cadherin protein in BTCC tissue.It may play a key role in progression of BTCC.
