骨转移瘤中侧群细胞的初步分析

摘　　要：目的研究骨转移瘤原代细胞中是否存在干细胞样侧群细胞(SP细胞)。方法对乳腺癌、非小细胞肺癌、前列腺癌、肾癌及宫颈癌骨转移瘤原代细胞进行Hoechst33342染色,应用荧光显微镜直接进行观察,检测骨转移瘤原代细胞中是否存在SP细胞及非SP细胞,并给予维拉帕米拮抗剂,观察各骨转移瘤瘤细胞中SP细胞亚群染色变化情况。结果所检测的乳腺癌、非小细胞肺癌、前列腺癌、肾癌及宫颈癌骨转移瘤原代细胞中均存在SP细胞。荧光显微镜下,SP细胞体积较非SP细胞小,SP细胞胞核蓝色荧光呈淡染或消失;非SP细胞经染色后发出较强的蓝色荧光。给予维拉帕米拮抗剂后,各骨转移瘤细胞中胞核发蓝色荧光的细胞数目明显增多。结论乳腺癌、非小细胞肺癌、前列腺癌、肾癌及宫颈癌骨转移瘤原代细胞中存在干细胞样的SP细胞。Objective To detect whether the side population cells exits in bone metastasis of malignant tumors.Methods In bone metastasis of breast cancer,no-small cell lung cancer,prostatic carcinoma,renal carcinoma and cervical cancer,stem-like cancer cells were identified using the fluorescence microscope and analysis of cells DNA stained with Hoechst33342.Results The SP cells and non-SP cells sorted by this method was observed by electron fluorescence microscope,and the change of blue area are observed after treated by Verapamil.SP cells were smaller than non-SP cells,and their blue fluorescence were weak or disappeared.Non-SP cells emit strong blue fluorescence.The blue area of bone metastasis tumors after treated by Verapamil increased.Conclusion The side population cells sorted from bone metastasis of malignant tumors are enriched with stem-like cancer cells.
