汉防己甲素逆转脑胶质瘤耐药性的实验研究

摘　　要：目的研究汉防己甲素对脑胶质瘤细胞耐药性的逆转作用。方法取人脑胶质瘤细胞株U251,在BCNU作用下建立耐药细胞株U251/BCNU,用MTT检测其耐药指数。在汉防己甲素(tetrandrine,TTD)作用后,检测其逆转倍数。结果 BCNU对U251和U251/BCNU细胞的半数抑制率(IC50)分别为12.00μg/ml,228.40μg/ml,耐药指数约为19。加入TTD后,BCNU对U251和U251/BCNU细胞的半数抑制率(IC50)分别为10.23μg/ml,22.87μg/ml,逆转倍数约为10倍。结论 TTD能逆转人脑胶质瘤细胞对BCNU的耐药性。Objective To investigate the reversal effects of tetrandrine on drug-resistance in glioma. Methods In vitro,BCNU-resistance human glioma U251/BCNU was established. By MTT assay,resistance index was detected. After exposing to tetrandrine,reversal fold was detected. Results In exposing to BCNU,the IC50 of U251 and U251/BCNU was 12.00 μg/ml and 228.40 μg/ml respectively. The resistance index was 19. After combined with tetrandrine,the IC50 of U251 and U251/BCNU was 10.23 μg/ml and 22.87 μg/ml respectively. The reversal fold was 10. Conclusions The drug-resistance against BCNU in glioma can be reversed by tetrandrine.
