摘  要:成纤维细胞生长因子21(FGF21)与其传统的FGFs家族不同,是一个新的代谢调节因子,虽然发现时间不长,但在代谢性疾病中的调控机制研究开展十分迅速。FGF21是2型糖尿病的潜在治疗剂,其具有持续控制脂质和糖代谢,逆转胰岛素的耐受性,改善胰岛β细胞等功能。另外,在特定靶组织/器官,很多与代谢相关的关键因子和途径,与FGF21调控机制相关。进一步研究这些作用机制不仅能更好的理解FGF21的生物学特性,还能为研究新的治疗方案奠定基础。文中叙述了糖尿病、肥胖症、肝癌、慢性肾病等疾病与FGF21的调控关系,以及目前FGF21调控机制的研究进展。Fibroblast growth factor 21(FGF21) is a new metabolic regulator,with several molecular characteristics that differ from classical FGFs,providing a variety of pharmacologic functions that including various metabolic responses in vitro and vivo in animal models.Although FGF21 was discovered relatively recently,the understanding of its biology and therapeutic utility is quickly evolving.FGF21 represents a potent therapeutic agent for obesity and type 2 diabetes,because of its ability to sustain lipid and gloucose control,it ameliorates insulin resistance,and improves β-cell function,without any apparent adverse effects.In addition,a large amount of crucial metabolically linked molecules and pathways have been considered to be involved in the mechanisms of action of FGF21,depending on the specific target tissue/organ.Further studies into these mechanisms should lead to more effective and important evidences to defining of FGF21 biology and pave the way for novel strategies in optimizing FGF21 therapeutic propertics.The specifics of FGF21 activities both in cell culture and in vivo,its potential as a new regulator for diabets,obesity,liver cancer and chronic renal disease,as well as the insighs into the molecular mechamisms of FGF21 metabolic actions will be discussed in this review.
