摘  要:目的:为研究角膜上皮下混浊(haze) 形成的发生机理,检测激光角膜光学切除术(photorefractive keratectomy ,PRK) 后角膜上皮和基质表达转移生长因子- α(transforming growth factor- α,TGF-α) 的变化。方法:新西兰白兔16 只,随机分成4 组,其中12 只施行PRK。于术后1 、2 、3 个月用裂隙灯显微镜观察haze 形成情况,并用原位核酸分子杂交方法,检测角膜上皮和基质TGF- αm RNA 的表达。结果:PRK 后1 个月已有haze 形成,2 个月haze 最明显,术后3 个月haze 减轻或消失。正常角膜上皮有TGF- αm RNA 表达,基质中未见其表达。PRK 后角膜上皮表达TGF- αm RNA 增加,且以术后1 、2 个月表达最明显。TGF- αm RNA 表达强弱与形成haze 的轻重相关。结论:TGF- α参与PRK 后伤口愈合过程,且调节着hazeIn order to study the mechanism of haze formation and investigate the expression changes of transforming growth factor-α(TGF-α) mRNA in corneal epithelium and stroma after photorefractive keratectomy (PRK),sixteen white rabbits were randomly divided into 4 groups,and PRK was performed on each eyes of 12 rabbits The haze formation was examined under a slit-lamp microscope at the 1st,2nd and 3rd month after PRK,and the expression of TGF-α mRNA was detected with in situ hybridization The cornea haze had formed at the 1st month after PRK The most obvious haze formation was observed at the 2nd month,and decreased and disappeared at the 3rd month after PRK TGF-α mRNA expression was present on the normal corneal epithelium and not on the corneal stroma The capacity for corneal epithelium expression of TGF-α mRNA increased after PRK,and the peak appeared on the 1st or 2nd month The extent for expression of TGF-α mRNA was relative to the haze formation TGF-α takes part in promoting corneal wound healing after PRK and may contribute to corneal haze formation and development
