摘 要:目的观察乙醇对新生小鼠延髓基本节律性呼吸的作用。方法制作新生小鼠离体延髓脑片标本,包含面神经后核内侧区并保留舌下神经根,给予灌流改良Kreb′s液记录舌下神经根的节律性呼吸放电活动(RRDA)。实验分为2组:模型验证组使用改良Kreb′s液灌流脑片标本,记录舌下神经根的RRDA60 min,观察舌下神经根的RRDA有无改变,以判断实验模型的稳定性;实验组使用含0 mg.L-1(对照组)、200 mg.L-1(200 mg.L-1乙醇组)、800 mg.L-1(800 mg.L-1乙醇组)乙醇的改良Kreb′s液灌流脑片,每个浓度灌流15 min,对比RRDA改变,分析乙醇对延髓基本节律性呼吸作用。结果模型验证组舌下神经根的RRDA的呼吸周期、吸气时程、放电积分幅度等呼吸指标在60 min变化无统计学差异;200 mg.L-1的乙醇缩短呼吸周期、延长吸气时程、增加放电积分幅度,对RRDA有兴奋作用;800 mg.L-1的乙醇延长呼吸周期、缩短吸气时程、降低放电积分幅度,对RRDA有抑制作用。结论低质量浓度的乙醇对新生小鼠延髓基本节律性呼吸有兴奋作用,高质量浓度的乙醇对其有抑制作用。Objective To investigate the effect of alcohol on respiratory-related rhythmic discharge activity(RRDA) of medulla oblongata in neonatal mice.Methods Experiments were performed on neonatal mice medullary slices in vitro.The slices containing the medial region of the nucleus retrofacialis with the hypoglossal nerve rootlets retained were prepared during which the RRDA was recorded from hypoglossal nerve rootlets by suction electrode.The possible effect of alcohol on RRDA was investigated by dissolving alcohol in modified Kreb′s solution(MKS),for perfusion of the brainstem slices.Twelve positive slices of RRDA were divided into 2 equal groups randomly:One was authenticated group,the slices were perfused with MKS;the other was experimental group,the slices were perfused with different concentrations(0 mg·L-1,200 mg·L-1,800 mg·L-1)of alcohol.Results The respiratory cycle,inspiratory time,integral amplitude of RRDA in authenticated group had no change in 60 min,the methods of this experiment was feasible.At the concentration of 200 mg·L-1,alcohol shor-tened respiratory cycle,prolonged inspiratory time,and enhanced integral amplitude,which exerted the exciting effect on RRDA.By contrast,if the concentration of 800 mg·L-1,alcohol produced opposite effect.Conclusions Alcohol in a low concentration has an exciting effect on RRDA of neonatal mice,and has a rejection capability effect on RRDA in a high concentration.
