生长抑素类似物奥曲肽对人胃癌生长的抑制作用研究

摘　　要：目的探讨生长抑素类似物奥曲肽对人胃癌生长的抑制作用。方法 将50例胃癌患者随机分为对照组(25例,术前不用药物)和奥曲肽组(25例,术前奥曲肽100μg皮下注射,1次/d,共7 d)。术中切除标本送组织学研究,分析胃癌组织坏死、炎性细胞浸润及纤维组织增生情况。用免疫组化法检测胃癌组织中p63蛋白及增殖细胞核抗原(PCNA)表达情况,用DNA末端原位标记染色法检测胃癌细胞凋亡情况。结果与对照组比较,奥曲肽组胃癌组织坏死、纤维组织增生明显(P<0.05),胃癌细胞凋亡率显著增高[(8.85±1.92)%与(6.47±1.68%)],p63蛋白及PCNA阳性表达率显著降低,差异均有统计学意义(P<0.05)。结论奥曲肽对人体内胃癌生长具有明显抑制作用。Objective To investigate the inhibitive effect of somatostatin（SST） analog octreotide on human gastric cancer. Methods Fifty gastrc cancer patients were randomly divided into groups control（n=25,without medicine before operation）,combination（n=25,received octreotide subcutaneously,100 μg/d,7 days before surgery）.The resected specimens were examined histologically,and tumor tissue necrosis,inflammatory cells infiltration and fibroustissue proliferation were analyzed.The expression of p63 protein and proliferating cell nuclear antigen in gastric cancer were evaluated by immunohistochemical methods and the apoptosis of tumor cells by terminal deoxynucleotidyl transferase mediated-dUTP nick end labeling. Results Gastric cancer tissue necrosis and fibroplasias were significantly higher in combination group than in control（P〈0.05）,and the apoptosis rate higher [（8.85±1.92%） vs（6.47±1.68%）]（P〈0.05）,but the expressions of p63 protein and PCNA lower（P〈0.05）. Conclusion Octreotide can inhibit the growth of human gastric cancer.
