摘  要:目的:探讨转化生长因子(TGF)-β1和白细胞介素(IL)-10在实验性自身免疫性重症肌无力(EAMG)发病机制中的作用。方法:10只Lewis大鼠随机分为EAMG组和健康对照组。EAMG组采用AChR二次免疫的方法制作EAMG模型,观察大鼠EAMG症状。于初次免疫后第7周,对大鼠进行Lennon评分,采用Western Blot和RT-PCR方法分别检测大鼠脾脏T淋巴细胞TGF-β1和IL-10的表达水平。结果:AChR二次免疫后,EAMG组大鼠产生了典型的EAMG症状。初次免疫后第7周,EAMG组大鼠Lennon评分及其脾脏T淋巴细胞TGF-β1和IL-10mRNA的表达水平均显著高于健康对照组(P﹤0.01)。结论:EAMG大鼠脾脏T淋巴细胞TGF-β1和IL-10的表达增强,提示TGF-β1和IL-10在EAMG的发病机制中具有重要作用。Objective:To investigate the effect of transforming growth factor(TGF)-β1 and interleukin(IL)-10 on the pathogenesis of experimental autoimmune myasthenia gravis(EAMG).Methods:10 Lewis rats were divided randomly into EAMG group and normal control group.The EAMG mode were induced with the methods of AChR for 2 times in EAMG group,then the symptoms of EAMG were viewed.After 7 weeks of the first immunization,the symptoms of rats were scored as Lennon,the expression of TGF-β1 and IL-10 of spleen T cells of rats were measured by the methods of Western Blot or RT-PCR.Results:The rats of EAMG group showed typical symptoms of EAMG after the second immunization,of which the Lennon score and the expression of TGF-β1 and IL-10 of spleen T cells were higher significantly than the normal control group after 7 weeks of the first immunization (P﹤0.01).Conclusions:The expression of TGF-β1 and IL-10 of spleen T cells of EAMG rats were enhanced,which suggested that TGF-β1 and IL-10 play an important role in the pathogenesis of EAMG
