摘  要:目的研究维生素E对H2O2引起的人甲状腺细胞凋亡的影响。方法取良性甲状腺腺瘤手术中的腺瘤旁正常甲状腺组织进行细胞培养。预防组加VE(50μmol/L)或无血清培养液后再加入不同浓度H2O(2200~800μmol/L),治疗组预先加入不同浓度H2O(2200~800μmol/L)后再加入VE或无血清培养液刺激单层培养的甲状腺细胞,MTT法检测细胞存活率,流式细胞术(FCM)检测各组甲状腺细胞凋亡率。结果预防组和治疗组单纯200~800μmol/L H2O2作用24 h均可使甲状腺上皮细胞存活率降低、凋亡率升高(P<0.01),并呈浓度效应关系。与相应单纯H2O2处理相比,加入50μmol/L VE预干预及治疗后,细胞存活率升高、凋亡率下降,400、800μmol/L H2O2处理时上述反应尤为显著(P<0.05)。结论维生素E对H2O2诱导的人甲状腺细胞凋亡具有保护作用。This study aimed to investigate the influence of vitamin E(VE) on apoptosis of human thyrocyte induced by H2O2.The human thyroid epithelium cells(TEC) were collected from normal para-adenoma tissues of patients underwent thyroid adenoma resection.In prevention groups,vitamin E(50 μmol/L) or serum-free medium was respectively added into thyrocytes of monolayer culture before H2O2 addition of different concentrations,while in treatment groups H2O2(200-800 μmol/L) was added firstly.The survival rates of thyrocyte were detected by MTT,while the apoptosis rates were detectd by flow cytometry.When the thyroid epithelium cells exposed to H2O2(200-800 μmol/L) for 24 hours,the cell survial rates declined and cell apoptosis rates increased(P 0.01) in both prevention and treatment groups,with a relation of dose-effect response.Adding VE(50 μmol/L) into cell culture for prevention and treatment,the cell survial rates increased and the thyrocyte apoptosis reduced,as compared with the corresponding simple H2O2 intervention groups.Above differences were particularly significant(P 0.05) when thyrocytes exposed to 400 and 800 μmol/L H2O2.Thus,we concluded VE has protective effects on H2O2-induced human thyrocytes apoptosis.
