摘  要:目的探讨β-catenin、cyclinD1在乳腺癌中的表达及意义。方法应用免疫组织化学SP法检测99例乳腺癌,56例乳腺增生症和32名正常乳腺组织中β-catenin、cyclinD1的表达,并分析其与乳腺癌临床病理特征的关系。结果 32名正常乳腺组织中β-catenin在细胞膜中强表达,cyclinD1阴性表达。乳腺癌β-catenin异常表达率78.8%,明显高于乳腺增生症42.8%(P<0.01);乳腺癌cyclinD1阳性表达率57.6%,高于乳腺增生症32.1%(P<0.01)。β-catenin异常表达、cyclinD1的过表达与淋巴结转移相关。β-catenin的异常表达与cyclinD1的表达相关。结论β-catenin异常表达是乳腺癌发生发展过程中的重要因子,这个过程可能是通过激活cy-clinD1实现的。β-catenin异常表达和cyclinD1过表达可作为评估乳腺癌转移预后的指标。Objective To study the expression and significance of β-catenin,cyclinD1 in breast carcinoma.Methods Ninety-nine breast carcinomas,56 breast cystic hyperplasia tissues and 32 normal breast tissues were studied.Immunohistochemistry was used to detect the expressions of β-catenin and cyclinD1,then their relationship with the clinicopathological features of breast carcinomas were analyzed.Results In 32 cases of normal breast tissues,epithelial cells showed equally strong membranous of β-catenin protein at the cell-cell boundaries,but the expression of cyclinD1 was negative.The abnormal rate of β-catenin in breast carcinomas(78.8%) was much higher than that of breast cystic hyperplasia tissues(42.8%)(P0.01).The overexpression of cyclinD1 in breast carcinomas(57.6%) was much higher than that of breast cystic hyperplasia tissues(32.1%)(P0.01).The abnormal expression of β-catenin and the overexpression of cyclinD1 was associated with lymph node metastasis.There was a highly significant association between abnormal expression of β-catenin and overexpression of cyclinD1.Conclusion The abnormal expression of β-catenin plays an important role in the carcinogenesis and progression of human breast carcinoma,the mechanism of which may be up-regulating the expression of cyclinD1.Abnormal expression of β-catenin and the overexpression of cyclinD1 could be considered as useful makers for determining metastasis and prognosis of human breast carcinoma.
