卵巢浆液性囊腺癌与交界性囊腺瘤的差异蛋白质组学分析

摘  要:目的探讨卵巢浆液性囊腺癌(CA)与交界性囊腺瘤(IN)的差异表达蛋白质谱,从而寻找特异性诊断标志物。方法采用二维液质联用分析技术。结果 CA-IN组鉴定得出的96个差异蛋白中,有83个差异蛋白表达上调;13个差异蛋白表达下调。结论初步构建起卵巢浆液性囊腺癌的差异蛋白质谱。差异蛋白中细胞骨架蛋白、伴侣蛋白、细胞外基质蛋白、信号转导、细胞周期、转录及翻译相关蛋白等,特别是细胞周期调控、转录及翻译调控蛋白对卵巢癌发生、发展、转移有重要意义。在卵巢浆液性囊腺癌中的表达异常为首次报道。这些蛋白将有利于卵巢浆液性囊腺癌及卵巢浆液性囊腺瘤的鉴别诊断。一些关键的差异蛋白如POSTN、SPON1等经过进一步临床验证后有望成为卵巢癌的肿瘤标志物,将对卵巢癌的发病机制阐述、诊断、治疗及预后评估有重要意义。Objective Ovarian serous cystadenocarcinoma(CA) and borderline cystadenoma(IN) of differentially expressed proteins,in order to find specific diagnostic markers.Methods Using two dimensional liquid chromatography MS/MS(2-D LC-MS|MS) technology.Results Identification of CA-IN groups drawn from 96 different proteins,there are 83 differences in protein expression;13 differential protein expression decreased.Conclusion Initially built ovarian serous cystadenocarcinoma protein spectrum of the difference.Cytoskeletal proteins,chaperone proteins,extracellular matrix proteins,signal transduction,cell cycle,transcription and translation-related protein and other differences in protein,especially in cell cycle regulation,transcription and translation regulatory proteins in ovarian cancer,development and transfer are important.In ovarian serous adenocarcinoma exception was first reported.These proteins will help to ovarian serous adenocarcinoma and ovarian serous cystadenoma of the differential diagnosis.Some of the key differences between proteins such as POSTN,SPON1 and so on through further clinical testing is expected to be after the tumor markers of ovarian cancer,will elaborate the pathogenesis of ovarian cancer,diagnosis,treatment and prognosis are important.
