摘  要:目的研究不同程度的能量限制(CR)下,大鼠脑组织中SIRT1和SIRT2的表达变化。方法 60只大鼠随机分成四组:正常对照组、75%CR组(喂养食物为正常对照组的75%),55%CR组(喂养食物为正常对照组的55%)和高脂组,喂养8周。免疫组织化学检测大鼠脑组织中SIRT1和SIRT2的表达与定位。RT-PCR及Western-blot检测各实验组中SIRT1、SIRT2的表达。结果在大鼠脑组织中发现SIRT1可于细胞浆和细胞核中表达,且主要于细胞核中表达;SIRT2主要在细胞核中表达。能量限制下,与对照组相比SIRT1表达升高;高脂食物条件下,SIRT1表达相对于对照组增高,但是比CR组减少。75%CR与55%CR相比,后者中的SIRT1的表达增加更多。与对照组相比,55%CR增加SIRT1的表达差异有统计学意义(P<0.05),而75%CR和HFa增加SIRT1的表达差异无统计学意义。SIRT2在CR和HFa下表达无明显变化。结论 CR能增加SIRT1而不增加SIRT2的表达,重度CR比中等程度CR对SIRT1表达的影响更大。Objective To investigate the effect of calorie restriction on the expression of SIRT1 and SIRT2 in rat brain.Methods Sixty healthy Sprague-Dawley rats were randomly divided into four groups:normal control(NC),75% calorie restriction(CR),55%CR,and high fat(HF) groups.The rats were maintained for 8 weeks.Immunohistochemical staining method was used to detect the expression and localization of SIRT1 and SIRT2 in the rat brain.RT-PCR and Western-blot methods were used to detect the expression of SIRT1 and SIRT2.Results SIRT1 was found to distribute more in the nucleus than in the cytoplasm,and SIRT2 was found only in the nucleus.After CR,the expression of SIRT1 was increased.For the HF group,the expression level of SIRT1 was higher than the control NC group.The expression of SIRT1 in the 55% CR group was significantly high than the 75%CR group(P0.05).The increase in the SIRT1 expression in the 75% CR and the HF groups was not significant.The expression level of SIRT2 was not changed in the rat brain.Conclusion CR can upregulate the expression of SIRT1,but not SIRT2;Extensive CR can significantly increase the expression of SIRT1 in rats brain.
