摘  要:目的观察载脂蛋白E基因敲除(ApoE KO)对小鼠海马神经元内β淀粉样蛋白(Aβ)表达的影响,探讨其与神经退行性疾病的认知功能障碍之间的可能联系。方法 10只正常C57BL/6J小鼠予普通饲料喂养作为对照(C)组,10只ApoE-KO的C57BL/6J小鼠予普通饲料喂养作为KO组,10只ApoE KO的C57BL/6J小鼠予高脂饲料喂养作为KO-HF组。小鼠成模后称重,取血检测血脂,取小鼠脑组织分别进行HE、免疫组化染色,并应用计算机图像分析系统检测小鼠海马神经元内Aβ的表达变化。结果与C组比较,KO组和KO-HF组体重、总胆固醇、甘油三酯及低密度脂蛋白胆固醇含量明显升高,差异有统计学意义(P<0.05),HE染色结果显示:与C组比较,KO组与KO-HF组小鼠海马神经元结构在光镜下无明显区别;免疫组化染色可见KO组及KO-HF组小鼠海马神经元内有黄色颗粒样Aβ沉淀;与C组比较,KO组平均光密度值升高,差异有统计学意义(P<0.05),与KO组比较,KO-HF组平均光密度值升高,差异有统计学意义(P<0.05)。结论 ApoE KO及高脂等因素会导致Aβ的沉积,与ApoE异常导致阿尔茨海默病等神经退行性疾病的认知功能障碍可能有着密切联系。To observe the effect of the knockout of ApoE gene( ApoE KO) on the expression of amyloid protein 13(Aβ) in the hippocampaI neurons of the mice and discover its possible relationship with Alzheimer Disease's pathologic processes. Methods Ten wild-type and ten ApoE KO mice were fed with common chow as the control group (group C) and the KO group respectively, while 10 ApoE KO mice were fed with high fat diet as group KO-HF. Twelve weeks later, the weight and the lid of these mice were measured. The brain tissues were observed by HE and immunohistochemistry staining. Then used the image analyzing system of the computer to detect the expression of Aβ in the hippocampal neurons of the mice. Results In the ApoE KO group, weight, total cholesterol, triglyceride ,low-density lipoprotein cholesterol were higher than those in the control group, and these changes were more significant in group KO-HF. The average optical densities of Aβ were (0. 239 5±0. 085 8), (0. 288 5± 0. 121 3) and (0. 350 7 ±0. 141 5) in group C, group KO and group KO-HF respectively, while these data were all significant(P 〈 0. 05 ). Conclusion The knockout of ApoE gene and high fat diet can lead to the deposition of Aβ which may have close relationship with the occurrence and development of Alzheimer Disease's pathologic processes.
