摘  要:目的探讨叶酰聚谷氨酸合成酶(FPGS)在氨甲蝶呤(MTX)对映体[L-(+)-MTX和D-(-)-MTX]耐药A549细胞株中的表达。方法通过FQ-PCR和Western blot法分别测定肺癌A549细胞株和15μmol/L L-(+)-MTX和D-(-)-MTX两种耐药A549细胞株中FPGS mRNA和蛋白表达。结果 L-(+)-MTX、D-(-)-MTX耐药细胞株中FPGS基因表达的mRNA相对含量分别为肺癌A549细胞株的(0.80±0.09)倍和(2.04±0.34)倍,两组间差异具有统计学意义(P<0.05);L-(+)-MTX、D-(-)-MTX耐药细胞株中FPGS的蛋白表达含量分别为对照组肺癌A549细胞株的(0.85±0.12)倍和(1.62±0.24)倍,两组间差异具有统计学意义(P<0.05)。结论 MTX诱导耐药后细胞株中FPGS mRNA和蛋白均发生变化,且在两种对映体细胞株间具有手性差异。Objective To study the expression of folylpolyglutamate synthetase (FPGS) in methotrexate (MTX) enantiomer-resistant A549 cell lines [ L-( + )-MTX and D-( - ) -MTX]. Methods The expression of FPGS on genetic and protein level was determined by FQ-PCR and Western blot in lung cancer A549 cells, and MTX enantiomer-resistant A549 cells [ L-( + )-MTX and D-( - )-MTX], with the concentration of drug resistance was 15 ixmol/L. Results The genetic expression level of FPGS was (0. 80 ± 0. 09 ) and ( 2. 04 ± 0. 34) folds in L- ( + ) - MTX/A549 cells and D-( - )-MTX/A549 cells compared with lung cancer A549 cells, there was statistical differ ence between two groups ( P 〈 0.05 ). The protein expression level of FPGS was (0. 85 ± 0. 12 ) and ( 1.62 ± 0. 24) folds in L-( + )-MTX/A549 cells and D-( - )-MTX/A549 cells compared with lung cancer A549 cells, there was statistical difference (P 〈 0. 05 ). Conclusion The expression level of FPGS on genetic and protein level in drug resistant cells have been changed, and significant difference in two enantiomer-resistant cells are appeared.
