摘  要:目的探讨电针对先天性脑损伤仔鼠脑组织脑源性神经生长因子(BDNF)表达的影响和机制。方法给孕17 d的脂多糖组孕鼠腹腔注射脂多糖450μg/(kg.d),制备脑损伤模型;对照组孕鼠腹腔注射等量生理盐水。脂多糖组仔鼠随机分为非干预组和电针组,电针组于7日龄开始干预,每日1次,干预至28日龄。选取百会、曲池及足三里为干预穴位。对照组、非干预组仔鼠均常规饲养。采用免疫组织化学方法检测1、14、21、28日龄仔鼠脑组织BDNF表达情况。结果 1日龄非干预组脑组织BDNF阳性细胞计数高于对照组,差异有统计学意义(P<0.01),此后两组间比较差异无统计学意义(P>0.05);电针组各日龄仔鼠脑组织BDNF阳性细胞计数高于非干预组和对照组,差异有统计学意义(P<0.01)。结论宫内感染可引起脑损伤仔鼠脑组织BDNF阳性表达增强;电针能持续上调脑损伤仔鼠脑组织BDNF阳性表达。Objective To investigate the effects of electroacupuncture on the expression of brain-derived neuotrophic factor(BDNF) in the brain tissues of the young offspring of rats with congenital brain damage.Methods Prepare the brain damage models by injecting LPS 450 μg/(kg·d)into abdominal cavity of pregnant rats on d17 and d18;injecting equally sterile NS into abdominal cavity of contrasting pregnant rats.The LPS young offspring of rats were randomly divided into the non-intervention group and the electroacupuncture group.The electroacupuncture group began intervention from d7 until d28,once a day.Select Baihui,Quchi and Zusanli as intervention points.The control group and the non-intervention group were given regular feeding.Immunohistochemistry was used to detect the expression of BDNF in d1,d14,d21 and d28.Results The expression of BDNF of the non-intervention group was higher than the control group(P0.01)in d1,and there was significant difference between the two groups.Compared with the non-intervention group and the control group,the expression of BDNF of the electroacupuncture group was higher(P0.05)in each period and the difference was significant(P0.01).Conclusions Intrauterine infection can cause the increase of the expression of BDNF in neonatal rats with brain damage.Electroacupuncture can persistently increase the expression of BDNF in neonatal rats with brain damage.
