摘  要:①目的从肺癌转移相关的3个候选基因片段(C1、L1、L2)中筛选出目的基因片段,并克隆其基因全长。②方法 3种肺癌细胞株及4种肺癌手术标本总RNA的提取;运用RT-PCR技术中,对肺癌转移相关基因C1、L1、L2片段进行验证,钓取目的片段;运用RACE(cDNA末端快速扩增)技术克隆出其基因全长序列,并登录Genbank,进行序列比对,确定克隆的基因性质。③结果在NCI-H446细胞和95-D细胞中未得到C1和L1基因目的大小片段,只有L2基因目的片段;从2种细胞株中均得到L2基因的3′端部分未知序列1169 bp;从NCI-H446细胞中得到L2基因的5′端未知序列178 bp,从95-D细胞中145bp。3′、L2基因序列与已知的ABCE1基因序列同源性100%。④结论从已知的3个片段C1、L1、L2中,成功筛选与肺癌转移相关基因L2即ABCE1基因。Objective Based on part of known cDNA sequences of the lung cancer metastasis related genes C1,L1 and L2,to test and to obtain full-length sequences of them.Methods Three kinds of lung cancer cell line and 4 kinds of lung cancer the extraction of total RNA surgical specimens;Using RT-PCR technology,of lung cancer metastasis associated genes C1,L1,L2 clips verification,fish for purpose extract;Using cDNA rapid expansion that(at) technology cloned its genetic sequences,and login Genbank sword-boat,sequence alignment,determine the gene cloning properties.Results There is no EST of C1,L1 cloned in NCI-H446 and 95-D cells;while amplification the 3′terminal of known cDNA sequence of L2 is 1169 bp in NCI-H446and 95-D,and 5′terminal of known cDNA sequence is 178 bp in NCI-H446,145bp in 95-D.BLAST results shows that the full-length sequence of3,L2 has a sequence homology with Homo sapiens ATP-binding cassette sub-family E(OABP) member 1(chromosome 4q21),with expect = 0.0 and identities =100%.Conclusion From a known 3 segments C1,L1 and L2,test with lung cancer screening ABCE1 metastasis associated genes L2 namely genes successful.
