喉癌喉咽癌P-GP定量分析及临床意义

摘　　要：应用流式细胞仪结合计算机辅助行数据、图像分析，用荧光标记抗体（ Ｍ Ｄ Ｒ１ Ｐ Ｅ， Ｃｌｏｎｅ Ｕ Ｉ Ｌ２）定量检测３４ 例喉癌、喉咽癌组织和１８ 例癌旁组织的 Ｐ Ｇ Ｐ含量，并与肿瘤病理、生物学特性进行比较分析，研究喉癌、喉咽癌细胞 Ｍ Ｄ Ｒ１ 基因产物 Ｐ Ｇ Ｐ的表达及临床意义。结果显示，喉癌、喉咽癌 Ｐ Ｇ Ｐ阳性表达率为 ５８８％ （２２／３４）； Ｐ Ｇ Ｐ表达与喉癌、喉咽癌患者的临床分期无关（ Ｐ＞ ００５），与病理组织学分级及有无颈淋巴结转移有关（ Ｐ＜ ００１）。提示 Ｐ Ｇ Ｐ检测有助于估计肿瘤的生物学特性； Ｐ Ｇ Ｐ有可能成为有效的预后指标。Quantitative analysis and statistical image analysis of P GP were made by Flow Cytometry and computer on 34 tumor tissues,18 tumor adjacent tissues in the patients with laryngeal and hypopharyngeal squamous cell carcinoma (LSCC,HSCC) and 3 normal laryngeal tissues,determined by MDR1 PE,cloneUIL2.Also the relationship between the expression of MDR1 and the clinical pathological characteristics of LSCC and HSCC were evaluated.The result revealed the rate of the positive expression of P GP was 58.8%(22/34).It also showed no correlation between P GP and clinical stages of tumors(P>0.05), but there was significant correlation between pathological degree and matastases of cervical lymph nodes (P<0.01). P GP expression is probably a biological marker of prognostic value to LSCC and HSCC.
