摘 要:目的探讨建立小鼠非酒精性脂肪肝动物模型的最佳饲料配比方案,为进一步开展脂肪肝及其治疗的相关研究提供有价值的实验数据。方法 80只KM小鼠随机分为4组,甲、乙、丙为模型组,丁组为对照组。模型组给予不同梯度的高脂饮食,对照组给予正常饮食。在造模后每1周分别取模型组动物各5只处死,观察其病理模型形成情况,做血液指标检测及病理切片;采用全自动生化分析仪检测生化指标;取肝脏,制肝匀浆,测定LTG、LTC。结果造模过程中,模型组动物乙组造模后体重增长较正常对照组及模型组甲组、丙组迅速。至实验结束时,甲组体重较正常组平均重7.62%,乙组体重较正常组平均重16.76%,丙组体重较正常组平均重11.14%,模型动物体重及肝指数均较正常组显著增加(P<0.01)。生化指标检测显示模型组小鼠的TC、TG、ALT、AST、LDL-C、LTG、LTC水平,均有一定程度的降低(P<0.01),HDL-C水平显著升高(P<0.01),其中乙组小鼠的变化幅度最大(P<0.01)。在病理方面,造模组1周后乙组小鼠最早出现了轻度的肝脂肪变,造模2周乙组小鼠多呈中度脂肪变。造模4周乙组小鼠呈中重度的大泡性脂肪变。结论适当的高脂高胆固醇配比饮食是一种快速有效的脂肪肝模型制作方法 。Objective Explore the establishment of non -alcoholic fatty liver in mice animal models of feed ratio of the best programs. Methods 80 KM mice were randomly divided into 4 groups A, B, C as a model group,D as the control group. Model group were given different gradient of high - fat diet, normal diet control group. Then in another week we collectod five each from the five model group randomly. And then killed them to observe the formation of pathological model, to do blood testing and do biopsy, and detected by biochemical analysis of serum biochemical indicators and the liver, liver homogenate system measured LTG, LTC. The results of the modeling process,the model group B increased the fastest than the otherss. Compared with the average weight of the normal, to the end of the experiment, group A were 7.62% heavier, group B were 16. 76%, group C were 11.14% ,the model animal weight and liver index increased significantly compared with the normal group (P 〈0. 01 ). Biochemical tests showed the model group mice TC, TG, ALT, AST, LDL - C, LTG, LTC level, there is a certain degree of reduction ( P 〈 0. 01 ) , HDL. - C levels were significantly increased ( P 〈 0. 01 ), where the sharpest change in group B mice (P 〈 0. 01 ). In pathology, after a week the group B first appeared in the mild hepatic steatosis, group B, 2 weeks after,mostly appear moderate fatty change. Modeling for g weeks in group B mice showed severe macrovesicular fatty change. Conclusion The ratio of the appropriate high - fat high - cholesterol diet is a fast and efficient production method of fatty liver model.
