摘  要:目的研究曲古抑菌素A(TSA)联合多西他赛(Doc)对肺腺癌细胞株A549凋亡和α-tubulin蛋白的影响。方法采用四甲基偶氮唑盐(MTT)比色法测定细胞抑制率,Hoechst33258染色法显示细胞凋亡,流式细胞术检测细胞周期的变化,免疫荧光及Western blot法检测Acetyl-α-tubulin蛋白表达的变化。结果 Doc对A549有明显的抑制作用,与TSA联合后凋亡率增加,与单独用药相比可发生明显的G2/M期阻滞(P<0.05),TSA联合Doc能够增加Acetyl-α-tubulin蛋白的表达。结论 TSA通过促使α-tubulin乙酰化水平增加Doc对A549的抑制作用。Objective To investigate the effect of administration of trichostatin A(TSA) and doeetaxel (Doe) on apoptosis and α-tubulin in lung adenocarcinoma A549 cells. Methods The cell inhibition ratio was evaluated using MTT assay. Apoptosis was assessed using Hoecbst 33258 staining and the cell cycle was detected by flow cytometry. Protein expression of Acetyl-α-tubulin was detected by immunofluorescence and Western blot. Results Doe significantly inhibited the growth of A549 ceils, and combined treatment with TSA induced severer apoptosis. The cell cycle was more markedly arrested in the G2/M phase compared with the single drug( P 〈 0.05 ). TSA and Doe increased protein expression of Acetyl-α-tubulin. Conclusion TSA induces sensitivity to Doe treatment in A549 cells through up-regulation of acetyl-ct-tubulin.
