摘  要:目的:观察戊四氮致痫大鼠脑神经型钙黏附素(N-cadherin)和神经营养因子-4(NT-4)表达的变化及灵芝孢子粉的干预治疗作用,研究癫痫的发病机制及灵芝孢子粉的作用机制。方法:健康雄性Wistar实验大鼠30只,随机分为正常对照组、癫痫模型组和灵芝孢子粉治疗组,每组10只。模型组和治疗组均用致痫亚惊厥剂量的戊四氮(PTZ)腹腔注射制作Wistar大鼠慢性点燃模型。在低温条件下迅速取脑,通过免疫组化和Westernblotting检测皮质和海马区N-cadherin和NT-4的变化。结果:实验性癫痫大鼠模型制作成功,灵芝孢子粉组和癫痫模型组实验动物均达到点燃标准,与癫痫模型组动物相比,灵芝孢子粉组大鼠潜伏期明显延长,但持续时间无显著差异。免疫组化和Western blotting检测实验结果表明:癫痫模型组实验大鼠脑海马和皮质N-cadherin含量比正常对照组增高(P<0.01);灵芝孢子粉组N-cadherins含量比癫痫模型组下降(P<0.01)。同时癫痫模型组大脑海马和皮质NT-4含量比正常对照组增加(P<0.05);灵芝孢子粉组NT-4含量比癫痫模型组增加(P<0.01)。结论:灵芝孢子粉能够降低N-cadherin的表达,调整病变神经元兴奋性,起到抗癫痫作用,还能增强NT-4的表达从而减轻癫痫发作对神经系统的损伤。AIM: To observe the effects of Ganoderma lucidum spore powder(GLSP) on the content of N-cadherin and neurotrophin-4(NT-4) in the brains of rats with epilepsy induced by pentetrazole(PTZ).METHODS: Thirty male Wistar rats were divided into 3 groups: GLSP group,epilepsy model group and control group(with 10 rats in every group).The epilepsy igniting model was induced by intraperitoneal injection of PTZ in the rats.The rats were killed and the brains were quickly collected at low temperature.The content of N-cadherin and NT-4 in cerebral cortex and hippocampus was measured with immunohistochemical staining and Western blotting.RESULTS: The rats in GLSP group and epilepsy model group all reached the "igniting" standard.Compared with epilepsy model group,the latent period in GLSP group extended obviously,but the duration was not obviously changed.Compared with control group,the content of N-cadherin and NT-4 in cortex and hippocampus increased obviously in epileptic model group.The content of N-cadherin was lower and the content of NT-4 was higher in GLSP group than those in epileptic model group(P0.01).CONCLUSION: Ganoderma lucidum spore powder attenuates the noxious injury by overexpression of N-cadherin and regulation of neuronal excitability in central nervous system in the rats,thus preventing the neural cells from the damages of epilepsy.Ganoderma lucidum spore powder also increases the expression of NT-4 to relieve seizure.
