摘 要:目的:建立肺癌性胸腔积液与结核性胸腔积液的双向电泳图谱,分析二者蛋白质表达的差异和特点。方法:以固相pH梯度等电聚焦为第一向,垂直板SDS-PAGE为第二向分离胸水蛋白质,银染显色,扫描仪获取凝胶图像并对其进行软件分析。应用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)鉴定差异蛋白质。结果:双向电泳结果显示,肺癌性胸腔积液中表达增高2倍的差异蛋白点52个,降低50%的蛋白质点60个。有9个点仅在结核性胸腔积液表达,11个点仅在肺癌胸腔积液中表达,并通过肽指纹图分析成功鉴定了6个蛋白质。结论:肺癌性胸腔积液组与结核性胸腔积液组的双向电泳结果显示蛋白表达图谱有明显差异,这些差异蛋白可能是肺癌相关蛋白或肺癌特异性蛋白标志物。Objective: To establish 2-dimensional electrophoresis profiles of the protein in human pleural effusion caused by lung cancer and pulmonary tuberculosis,and to analyze the difference in proteins expression between these two types of pleural effusion.Methods: The total proteins in pleural effusion were separated by immobilized pH gradient 2-dimensional gel electrophoresis(2-DE).After silver staining,the differential protein expression was analyzed using PDQuest analysis software.The differentially expressed proteins were analyzed and identified by matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALD I-TOF-MS).Results: The result of two-dimensional electrophoresis showed that 52 protein spots were highly expressed for two folds,and 60 protein spots were poorly expressed for 0.5 folds in pleural effusion caused by lung cancer;9 protein spots were expressed only in the pleural effusion caused by pulmonary tuberculosis;11 protein spots were expressed only in the pleural effusion caused by lung cancer.Six differential proteins were identified by peptide mass finger printing(PMF).Conclusion: There are significant differ-ences between pleural effusion protein expression of lung cancer and pulmonary tuberculosis,which may be lung cancer associated proteins or special protein mark.
