摘  要:将通过戊二醛交联得到的酶细胞共交联物截留于中空纤维膜反应器中,研究了催化剂负载量与膜透量和表观酶活以及比活的关系.考察了固定化酶细胞交联物膜反应器裂解青霉素钾盐(PGK) 的反应特征和使用寿命.结果表明,在内径15 m m 、有效长度175 m m 的膜反应器内装膜150根、膜有效表面积660 cm 2 、负载5 .0 g 的酶细胞交联物时,裂解25 批8 % 的PGK 可在90 min 内使其裂解率达到96 % .此结果显示,采用这种截留有酶细胞共交联的膜生物反应器裂解PGK 有希望成为目前工业酶法生产6 - APA 的可选技术.Cocross-linked enzyme-cell was held-back in hollow fiber membrane reactor after enzyme and cells were cross-linked together with glutaraldehyde. The relations between catalyst loading and membrane flux, and its apparent enzyme activity and its apecific activity were studied. The reaction character of cocross-linked enzyme-cell hydrolyzing PGK and the expectancy of catalyst were investigated when cocross-linked enzyme-cell was held- back in membrane. The results showed that 25 batches of 8% PGK were hydrolyzed by the catalyst within 90?min, the conversion was up to 96%, when 5.0?g cocross-linked enzyme-cell was held- back in membane reactor with 15?mm inner diameter and 175?mm effective length, which contained 150 membranes and the effective surface area of membrane was 660?cm 2. The results provided this immobilized catalyst which was obtained by cross-linking enzyme and cells together for becoming competitive power with the current industrial enzymatic producing 6-APA when it was held-back in membrane reactor.
