摘  要:目的探讨辛伐他汀对蛛网膜下腔出血大鼠海马肿瘤坏死因子-α(TNF-α)的影响。方法将SD大鼠分为对照组、模型组和治疗组,采用非开颅血管内穿线法制备大鼠SAH模型,治疗组给予辛伐他汀。采用免疫组织化学和免疫印迹方法检测大鼠海马TNF-α表达。结果模型组神经功能缺损明显,海马TNF-α表达明显高于对照组(P<0.05)。使用辛伐他汀后,TNF-α表达降低(P<0.05)。结论 TNF-α参与了蛛网膜下腔出血发病过程,辛伐他汀可能通过影响TNF-α途径发挥治疗蛛网膜下腔出血脑血管痉挛作用。Objective To observe the effect of simvastatin on tumor necrosis factor-alpha(TNF-α) in hippocampus of subarachnoid hemorrhage(SAH) model rats.Methods Thirty Sprague Dawley(SD) rats were divided into three groups randomly.Control group:only cistema magna puncture was done with no injection of blood.Model group:SAH model by endovascular perforation without opening cranium,Treatment group: rats were subcutaneously injected daily with simvastatin(20mg/kg,0.1ml) for 7 days after the first puncture.Expression of TNF-α protein in hippocampus were detected by immunohistochemistry and western-blotting.Results Compared with the control group,increased expression of TNF-α was detected in model group(P0.05).While lower expression of TNF-αwas detected in treatment group(P0.05).Conclusion Simvastatin treatment after SAH attenuated neurological deficits in rat.The mechanism may be attributable partly to TNF-α down regulation.
