肝细胞肝癌中Galectin-9 mRNA表达及其临床意义

摘　　要：目的:探讨Galectin-9 mRNA在肝细胞肝癌(简称肝癌)中的表达及其临床意义。方法:用TRIZOL法从89例肝癌患者手术切除的肝癌组织及癌旁组织中提取总RNA并反转录成cDNA,荧光定量PCR检测其Galetin-9 mRNA的表达量。结果:癌组织中的Galectin-9 mRNA表达较癌旁组织表达下降,仅为癌旁组织的0.504倍,两者比较差异有统计学意义(P<0.001);癌组织中的Galecin-9 mRNA表达的程度与肿瘤的大小、术前转移、病理分化、TNM分级、术后复发存在相关性(P<0.05);而与性别、年龄、HBsAg、AFP、肝硬化、包膜完整性无相关性(P>0.05)。结论:Galectin-9 mRNA的表达水平与肝癌的病理分化、转移、复发有关,可能成为诊断肝癌及判断预后的新生物学指标或治疗肝癌的潜在靶点。Objective：To investigate the expression of Galectin-9 mRNA in hepatocellular carcinoma （HCC） and its clinical significance. Methods：The tolal RNA was extracted from resectable cancer tissues and adjacent tissues in 89 patients with HCC, and reverse transcripted into cDNA, The expression of Galectin-9 mRNA was detected by the Real-time fluore^scence quantitative PCR. Results：The expression of Galectin-9 mRNA in tumor tissues was 0. 504-fold,lower than that in adjacent tissues （ P 〈0. 001）, and the level of Galecin-9mRNA expression in patients with HCC was assotiated with tumor size, preoperative metastasis, histological differentiation, TNM classification and recurrence （ P 〈0. 05 ）, but it was not related with gender, age, HBsAg, AFP, liver cirrhosis and the integrity of live capsule （P 〉0.05）. Conclusion：The expression of Galectin-9 mRNA in HCC is assotiated with histological differentiation, recurrence and metastasis, and maybe become the diagnostic and prognosis biological marker or potential new therapeutic target for HCC.
