卵巢上皮性肿瘤组织Nectin-4表达临床意义的探讨

摘　　要：目的:探讨卵巢上皮性癌(卵巢癌)组织中Nectin-4 mRNA和蛋白的表达及其与临床病理参数之间的关系。方法:采用逆转录聚合酶链式反应(RT-PCR)技术,检测30例卵巢癌、25例卵巢上皮性肿瘤和30例正常卵巢组织中Nectin-4 mRNA的表达情况;采用免疫组织化学法检测Nectin-4蛋白的表达情况,并分析其与卵巢癌临床病理参数的关系。结果:卵巢癌组Nectin-4 mRNA表达水平为0.71±0.12,卵巢良性肿瘤组为0.32±0.09,正常卵巢组为0.30±0.09,卵巢癌组的mRNA表达量显著高于后两组,差异有统计学意义,P<0.01。卵巢癌组Nectin-4蛋白表达水平为3.35±0.52,卵巢良性上皮性肿瘤组1.36±0.41,正常卵巢组1.15±0.44,卵巢癌组织的Nectin-4蛋白表达量明显高于卵巢良性上皮性肿瘤和正常卵巢组织,差异有统计学意义,P<0.01。卵巢癌组织中Nectin-4 mRNA和蛋白表达水平,在不同年龄、不同病理类型间比较,差异均无统计学意义,P>0.05;而在不同病理分化程度、手术分期及有无淋巴结转移间比较,差异均有统计学意义,P<0.05。结论:Nectin-4 mRNA和蛋白在卵巢癌组织中均呈高表达,而在卵巢良性肿瘤组织和正常卵巢组织中仅有微量表达。在卵巢癌组织中,其表达程度与病理分化程度、手术病理分期及淋巴结转移有密切关系。OBJECTIVE： To study the expression of the Nectin-4 mRNA and protein in ovarian cancer tissues, and investigate the relationship between the expression of Nectin-4 in epithelial ovarian cancer with the clinicopathologieal parameters. METHODS： The expression levels of Neetin-4 gene was evaluated by using RT-PCR and immunohistochemistry in normal ovarian tissues, ovarian benign tumor tissues and ovarian cancer tissues. And the relationship between the Nectin-4 and the clinicopathological parameters of ovarian cancer was analyzed. RESULTS： The expression of Neetin-4 mRNA was observed in 0. 71±0.12,0. 32±0. 09 and 0. 30±0. 09 of malignant ovarian tumors, benign ovarian tumors and normal ovarian tissue respectively, the expression of Nectin-4 protein was observed in 3.35±0. 52, 1. 36 ± 0. 41,1. 15 ±0. 44 of malignant ovarian tumors, benign ovarian tumors and normal ovarian tissue respectively. The expressions of Nectin-4 mRNA and protein in ovarian cancer were significantly higher than them in normal ovarian tissues and benign tumors（P〈0. 0l）. Statistical analysis revealed that the expression of Nectin-4 gene was not related to ages and histological subtypes（P〈0.05）, which was related to differentiation, operation staging and lymph metastases （P〈 0.01）. CONCLUSIONS： Nectin-4 is highly expressed and positively correlated with ovarian cancer, but it is expressed in normal ovarian tissues and ovarian benign tumor gleamLy. The changes of the expression of Nectin-4 gene is correlated with tumor differentiation degree, Surgical-pathological staging and the metastasis of lymph node.
