PDGF-BB对Fadu细胞增殖侵袭的影响及可能的机制研究

摘　　要：目的:研究不同浓度血小板衍生生长因子PDGF-BB诱导后Fadu细胞增殖侵袭能力及粘着斑激酶(FAK)、基质金属蛋白酶-2(MMP2)、基质金属蛋白酶-9(MMP9)表达的变化。方法:不同浓度(0,1,10,20,100 ng/mL)PDGF-BB诱导人下咽癌Fadu细胞株,RT-PCR方法检测FAK及MMP9、MMP2mRNA表达变化,噻唑蓝法(MTT)检测细胞增殖,侵袭实验检测细胞侵袭能力,Western blot检测不同浓度PDGF-BB处理后Fadu细胞蛋白表达变化。结果:诱导后细胞FAK、MMP9、MMP2mRNA水平上调,在20 ng/mL或10 ng/mL浓度时达到最高(与对照组相比P<0.05);各组蛋白表达增加与mRNA基本一致,在20 ng/mL达高峰(与对照组相比P<0.05),MTT示各组增殖无明显变化(与对照组相比P>0.05)。导后侵袭细胞数在各组均有增加,在20 ng/mL组达高峰为(20.12±4.5)个,较对照组差异有统计学意义(P<0.05)。结论:PDGF-BB上调Fadu细胞中FAK、MMP9及MMP2表达,从而使Fadu细胞侵袭能力增强,但对Fadu细胞增殖无明显影响。Objective： To investigate the dynamic changes in the expression of focal adhesion kinase （ FAK ）, matrix metalloproteinase-2 （ MMP-2 ）, and matrix metalloproteinase 9 （ MMP-9 in Fadu cells after stimulation with different concentrations of plate- let-derived growth factor-BB （PDGF-BB） and to explore their effect on cell proliferation and invasion. Methods： After Fadu cells are stimulated with different PDGF-BB concentrations, the dynamic changes in FAK, MMP-9, and MMP-2 mRNA expression in the cells were determined by reverse transcriptase polymerase chain reaction, and the changes in cell proliferation and invasive ability were determined with an MTT assay and a Transwell system assay, respectively. Western blot analysis was used for the protein expression. Results： The expression of FAK, MMP-9, and MMP-2 mRNA in Fadu cells was upregulated after stimulation with PDGF-BB and they reached their peak under 20 or 10 ng/mL of PDGF-BB （ P 〈 0.05 ）. FAK, MMP-9, and MMP-2 protein expression had the similar changes and all reached their peak in the 20 ng/mL group. Cell proliferation was not changed after induction. The number of invasive cells increased and reached their peaks in the 20 ng/mL group （ 20.12 ± 4.5 ）, which were significantly different from that in the control group （ P 〈 0.05 ）. Conclusion： PDGF-BB upregulates the expression of FAK, MMP-9, and MMP-2, which facilitates the invasion of Fadu cells, but PDGF-BB has no effect on their proliferation.
