维吾尔族宫颈癌人乳头瘤病毒感染与CD4^+CD25^+CD127^-调节性T细胞的相关性研究

摘　　要：目的:探讨维吾尔族宫颈癌人乳头瘤病毒(HPV)感染与CD4+CD25+CD127-调节性T细胞(Treg)的相关性。方法:采用HPV核酸扩增分型检测试剂盒及流式细胞术检测66例宫颈癌、35例宫颈上皮内瘤变(CIN)Ⅱ~Ⅲ、45例宫颈CINⅠ维吾尔族患者和40例维吾尔族正常对照组HPV分型和外周血中T细胞亚群及CD4+CD25+CD127-Treg的百分率。结果:①HPV感染率随着宫颈病变的加重呈趋势性增加。②外周血T细胞亚群在各组间比较,差异无统计学意义(P>0.05)。③宫颈癌组与宫颈CINⅡ~Ⅲ组患者外周血中CD4+CD25+CD127-Treg的百分率与宫颈CINⅠ组及正常对照组比较,差异均有统计学意义(P<0.05)。④HPV与CD4+CD25+CD127-Treg细胞呈显著正相关(r=0.560,P<0.05)。结论:Treg细胞可能是维吾尔族宫颈癌免疫逃避的机制之一,维吾尔族宫颈癌HPV与Treg细胞密切相关。Objective：To investigate the correlation between human papilloma virus （HPV） and CD4＋CD25＋CD127-T cells （Treg） of Uygurs.Methods：66 patients of Uygurs with cervical cancer,35 patients of Uygurs with cervical intraepithelial neoplasia （CIN）Ⅱ～Ⅲ,45 patients of Uygurs with CINⅠ and 40 normal controls of Uygurs were included in this study.Typing of HPV was evaluated by Nucleic acid amplification typing kit .The percentage of CD4＋CD25＋CD127lo/- T cells population in CD4＋ T cells was evaluated by flow cytometric analysis.Results：①Infection rate of HPV were tendency increased following aggravation of cervical lesion.②T cell subsets were no statistics difference in every groups.③The percentage of CD4＋CD25＋CD127lo/- T cells of CD4＋ T lymphocytes in peripheral blood from patients with cervical cancer and CINⅡ～Ⅲ were significantly difference in comparison with that in patients with CINⅠ and in normal controls （P0.05）.④ CD4＋CD25＋CD127-Treg was positive correlated with HPV（r=0.560,P0.05）.Conclusions：Treg may be one of the mechanisms for immune escape in cervical cancer patients of Uygurs,HPV is intimate correlated with Treg in cervical cancer of Uygurs.
