摘 要:目的:探讨STYK1/NOK基因对肺腺癌细胞迁移和侵袭的影响。方法:运用电穿孔仪将含STYK1/NOK全长基因的重组真核表达质粒(pcDNA3.1/NOK)转染入肺腺癌细胞系A549,筛选出稳定表达NOK蛋白的单克隆株A549-NOK。蛋白质印迹法检测转染前后细胞内NOK蛋白的表达效果。划痕实验、Transwell迁移实验检测转染前后细胞的迁移能力,Matrigel侵袭实验检测细胞的侵袭能力,MTT法绘制细胞生长曲线。结果:蛋白质印迹法显示,A549-NOK细胞中NOK蛋白表达量明显增高。划痕实验结果显示,A549-NOK细胞的迁移速度加快。在Transwell迁移实验中,A549-NOK组的穿膜细胞数较A549及空质粒对照组增多(95.7±9.5vs69.1±8.3、70.5±7.5),P值分别为0.003和0.004。Matrigel侵袭实验中A549-NOK组的穿膜细胞数较A549及空质粒对照组增多(67.8±6.7vs36.0±4.1、38.3±5.1),P值分别为0.000和0.001。MTT结果显示,A549-NOK细胞较A549和空质粒对照组细胞生长曲线上移。结论:STYK1/NOK基因能够促进肺腺癌细胞的迁移和侵袭,可能在肺癌的转移中发挥重要作用。OBJECTIVE: To investigate the impact of STYK1/NOK gene on the migration and invasion of lung adenocarcinoma cell line. METHODS: The recombinant STYK1/NOK eukaryotic expression plasmid was transfected into A549 cells with electroporation apparatus. The cloned strain A549-NOK with stable expression of NOK was screened, then the protein NOK was detected by Western blot. Cell migration was tested by scratch-wound assay and transwell migration assay. Cell invasion was tested by matrigel invasion assay. The cell growth curve was determined by MTT. RESULTS: Western blot showed the high expression level of NOK in A549-NOK cells. Scratch-wound assay showed the migrating speed of A549-NOK was faster than those of the controls. Transwell migration assay showed that the number of A549-NOK cells that passed the Transwell membrane was more than those of A549 and blank control cells (95.7±9.5 vs 69.1±8.3, 70.5±7.5, and P values were 0. 003 and 0. 004). Mattrigel invasion assay showed that the number of A549-NOK cells that passed the Transwell membrane was more than those of A549 and blank control cells (67.8±6.7 vs 36.0±4.1, 38.3±5.1, and P values were 0. 000 and 0. 001). The growth curve of A549-NOK cells was steeper than that of A549 and blank control cells.
