紫外分光光度法测定甲壳素的脱乙酰度

摘　　要：以0.1m ol/L盐酸为溶剂,以N- 乙酰- D- 葡糖胺(Ⅰ)和D- 葡糖胺盐酸盐(Ⅱ)为对照品,利用紫外分光光度法测定甲壳素的脱乙酰度.样品浓度在 440μg/m l-1100μg/m l范围内,线性关系良好(r= 0.9997),4 个不同批号样品测定的RSD 由0.09% ～1.9% (n= 5)与碱量法测定结果相符合.方法简便、快速。An UV spectrophotometry method for determination of degree of deacetylation of chitin using 0.1mol/L HCl as solvent and using N -acetyl- D -glucosamine and D -glucosamine as reference substance. Concentration of samples ranged from 440～1100μg/ml gave a good linear relationship ( r =0.9997). RSD of determination for samples No1-4 from 0.09% to 1.9%, Results of Determination agree with those that obtained by alkalimetric titration. The method is simple, rapid and accurate.
