摘 要:目的探讨脑肿瘤干细胞(BTSC)增殖活性与微血管密度的相关性。方法采用免疫组织化学染色检测BTSC标记物CD133、增殖细胞核标记物Ki-67和血管内皮细胞标记物CD31在胶质瘤组织中的表达;采用免疫荧光双染法检测CD133/Ki-67和CD133/CD31的共表达情况;并对BTSC的增殖活性与微血管密度的相关性行统计学分析。结果免疫组化显示:低级别胶质瘤中,CD133、CD31和Ki-67的阳性表达较少;而在高级别胶质瘤中,CD133、CD31和Ki-67阳性表达明显增加。免疫荧光双染显示:随着胶质瘤级别升高,CD133/Ki-67共表达明显增加,且可见CD133/CD31共表达于血管内皮细胞。在胶质瘤组织中,Ki-67、CD31表达均与CD133表达呈正相关(r1=0.758,P1=0.000;r2=0.439,P2=0.000),Ki-67与CD31的表达也呈正相关(r3=0.816,P3=0.000)。结论 BTSC与微血管联系密切,不仅在区域分布上围绕微血管增殖分化,并且在功能上与微血管相互促进和依赖。Objective To research the correlation between the proliferative activity of brain tumor stem cells(BTSC) and microvessel density.Methods The expressions of Ki-67,CD31 and CD133 were detected by immunohistochemistry,while the coexpressions of CD133/CD31 and CD133/Ki-67 were detected by immunofluorescence staining.Then,the correlation between the proliferative activity of BTSC and microvessel density was analyzed by statistical method.Results The positive expression of CD133,CD31 and Ki-67 was lower in low-grade glioma than in high-grade glioma by immunohistochemistry.Immunofluorescence staining showed that the coexpression of CD133/Ki-67 obviously increased with the rise of glioma grade.While CD133/CD31 coexpressed in vascular endothelial cell.In addition,there was a positive correlation between the expression of Ki-67,CD31 and CD133 in gliomas(r1=0.758,P1=0.000;r2=0.439,P2=0.000),and also a positive correlation between the expressions of Ki-67 and CD31(r3=0.816,P3=0.000).Conclusions In gliomas,the BTSC and microvessel are closely related to each other not only in the regional distribution but also in biological function.
